Embodiment 1

[0023] The acquisition of embodiment 1 variant attenuated strain of porcine epidemic diarrhea virus (abbreviated PEDV AJ1102-R) strain

[0024]PEDV AJ1102 strain (documentation: Jing Bi, a, b Songlin Zeng, a Shaobo Xiao, a Huanchun Chen, a and Liurong Fanga, Complete Genome Sequence of Porcine Epidemic Diarrhea Virus Strain AJ1102Isolated from a Suckling Piglet with Acute Diarrhea in China, GENOME ANNOUNCEMENT, Journal of Virology p.10910–10911) The virulently adapted Vero cell line. The attenuated strain with stable proliferation performance was obtained through cell subculture, plaque cloning and cultivation; the 96th passage virus was used to carry out 5 reversion tests in pigs, and it did not cause disease to 3-5 day old piglets, which proved the virulence of the passaged attenuated virus is stable. The applicant named the obtained attenuated strain as porcine epidemic diarrhea virus AJ1102-R strain, Porcine epidemic diarrhea virus AJ1102-R, and sent it to China. Wuhan. W...

Embodiment 2

[0042] The safety test of embodiment 2 attenuated strains of the present invention

[0043] 1. Pathogenicity test of different generations of PEDV

[0044] 1.1 Test method: The 15th generation, 35th generation, 55th generation, 75th generation, 96th generation, and 125th generation of PEDV were respectively orally infected with PEDV neutralizing antibodies [using Kusanagi et al. (Isolation and serial propagation of porcine epidemic diarrhea virus infection in cell cultures and Partial characterization of the isolate.J.Vet.Med.Sci.54,303–318) reported the improved neutralizing antibody detection method, the level of neutralizing antibody in the serum] is not higher than 3~4 sows produced by 1:4 5-day-old piglets, 5 piglets in each group, 1ml / head, and the challenge dose of the 15th generation was 10 6 . 5 TCID 50 , and the attack doses of the other generations were 10 7 . 0 TCID 50 , and set up another control group.

[0063] The protective efficacy test of embodiment 3 attenuated virus strain (PEDV) of the present invention

[0064] 1. Test material

[0065] 1.1 Viruses and vaccines

[0066] The virulent strain of porcine epidemic diarrhea virus AJ1102 was preserved and supplied by the Laboratory of Preventive Veterinary Medicine, School of Veterinary Medicine, Huazhong Agricultural University.

[0067] Three batches of porcine transmissible gastroenteritis and porcine epidemic diarrhea dual live vaccines (WH-1R strain + AJ1102-R strain) were trial-produced by the Preventive Veterinary Laboratory of the School of Veterinary Medicine, Huazhong Agricultural University.

Abstract

The invention belongs to the technical field of animal virology and animal-borne disease, and particularly relates to a low virulent strain variated from the porcine epidemic diarrhea virus and application thereof. The low virulent strain is a porcine epidemic diarrhea virus AJ1102-R strain having the preservation number of CCTCC NO:V201433, and has stable breeding performance after verification of cell passage and plaque cloning and culturing. The genomic sequence of the low virulent strain has the following variations: the 96th generation has sequence variation on the 71-72 of 3'UTR, and 2 bases TT are lost; and the 96th generation has sequence variation on the 3805-3816 of the S gene, and 12 bases GATGCTATTGTT are added. The immune efficiency of the low virulent strain can be used for inducing an immunized piglet to generate high-level neutralizing antibody and enough attacking protection, and can be used as a vaccine strain for application.

Description

technical field [0001] The invention belongs to the technical field of animal virology and animal infectious disease. It specifically relates to a variant attenuated strain of porcine epidemic diarrhea virus and its application. Background technique [0002] Porcine epidemic diarrhea (PED) is a highly contagious disease of pigs caused by porcine epidemic diarrhea virus (PEDV). The main features of the disease are watery diarrhea, vomiting and dehydration. Pigs of all ages are susceptible, and the incidence rate of 1-2 week-old suckling piglets is as high as 90%-100%, which brings great economic losses to the pig industry. In 1971, the occurrence of PED was first reported in the United Kingdom, and many countries such as Belgium, Germany, Switzerland, and Japan successively reported the occurrence of the disease (Pensaert M B et al, A new coronavirus-like particles associated with diarrhea in swine. Arch Virol, 1978 , 58:243-247; Takahashi K et al, An outbreak of swine diar...
