Embodiment 1

[0037] (1) Microwave extraction of copper algae polysaccharide: wash copper algae, control water, dry in an oven at 60°C, and crush. Take algae powder and add 95% ethanol solution to soak and degrease, and let stand overnight. Take 200g of copper algae powder, the ratio of solid to liquid is 1:20, the microwave time is 20min, the microwave power is 500W, and the temperature is 65°C for microwave extraction. At the end of the extraction, cool and stand for 1 hour, then refrigerate and centrifuge for 10 minutes (rotating speed 10000 rpm, temperature 4°C). The supernatant, that is, the crude polysaccharide extract was concentrated by rotary evaporation to 1-3 L, and 95% ethanol solution was added until the final volume fraction of ethanol was 30%, and stood overnight. The sample solution was refrigerated and centrifuged for 10 minutes (rotating speed 810,000 rpm, temperature 4°C), and the precipitate was collected, and a small amount of distilled water was added to dissolve the ...

Embodiment 2

[0050] The steps are the same as in Example 1, except that the time of enzymatic hydrolysis in step (3) in Example 1 can be fine-tuned to obtain polysaccharides with different molecular weight ranges.

Abstract

The invention discloses an enzymolysis-modified sargassum horneri polysaccharide and an application thereof. A preparation method of the enzymolysis-modified sargassum horneri polysaccharide comprises steps as follows: (1) performing microwave extraction on a sargassum horneri polysaccharide to obtain a coarse polysaccharide extract; (2) performing grading alcohol precipitation: performing precipitation on the coarse polysaccharide extract with the final ethanol volume fraction of 30-40% and 55-65% successively to obtain an SHP60 coarse polysaccharide component; (3) performing decolorization and deproteinization on the SHP60 coarse polysaccharide component to obtain an SHPA polysaccharide; (4) performing enzymolysis on the SHPA polysaccharide with a dextranase to obtain an enzymatic product; (5) performing ion column chromatography on the enzymatic product with a DEAE-Sepharose Fast Flow ion column chromatography system, and concentrating and filtering an eluent to obtain a filtrate; (6) performing gel column chromatography on the filtrate obtained in Step (5) with a Sephadex G-25 gel chromatography system, collecting a salt-free polysaccharide part, and performing concentration and freeze-drying to obtain the enzymolysis-modified sargassum horneri polysaccharide. The enzymolysis-modified sargassum horneri polysaccharide can be applied to preparation of an antioxidant, a moisture absorbent and a moisturizer.

Description

(1) Technical field [0001] The invention relates to an enzymatically modified copper algae polysaccharide and its application in the preparation of antioxidants, hygroscopic agents and moisturizing agents. (2) Background technology [0002] Polysaccharides are carbohydrates with complex structures and huge molecular weights, usually formed by dehydration and condensation of multiple monosaccharides. Polysaccharides in nature are widely distributed in organisms and are important components of living organisms. Seaweed polysaccharide has a complex structure and significant and diverse biological activities: its anti-oxidation and anti-aging effects are due to the use of seaweed polysaccharides to inhibit the activity of free radicals; the effects of promoting immunity and anti-tumor are mainly through improving the phagocytic ability of macrophages The effect of lowering blood sugar and blood fat is to utilize the adsorption of polysaccharides; and the anticoagulant effect is...
