Abstract

The invention relates to a method for preparing bitter gourd total saponins by an ultrahigh pressure assisted method and a purification method thereof. Reagents adopted by the method are free of volatility and toxicity, and saponin substances with high purity can be efficiently obtained in an environment-friendly mode. The method disclosed by the invention comprises the following steps: 1) cleaning, dicing, drying and crushing bitter gourds, and then sieving with a 60-mesh sieve to prepare bitter gourd powder; 2) weighing bitter gourd powder, and carrying out ultrahigh pressure extraction with an ethanol solution to obtain a bitter gourd total saponin extracting solution; (3) centrifuging the extracting solution, collecting supernate, carrying out rotary evaporation under the conditions of 40 DEG C and the rotating speed of 90r / min until no ethanol smell exists, and freeze-drying to obtain bitter gourd total saponin powder; 4) weighing the crude extract bitter gourd total saponin powder, and dissolving the crude extract bitter gourd total saponin powder in a 1-butyl-3-methylimidazolium tetrafluoroborate solution according to a material-liquid ratio; 5) fully stirring, adding a NaOH solution, and carrying out vortex oscillation until two phases are formed; 6) centrifuging under the conditions of 4 DEG C and 4000 r / min until two phases are fully separated; and 7) collecting a NaOH lower phase rich in saponin, neutralizing with hydrochloric acid, and freeze-drying to obtain the total saponin.
