The method comprises the following steps of: rinsing a jellyfish, cutting the jellyfish open from the top end of an umbrella part, and digging out mesoglea jelly; homogenizing the jelly, and putting homogenate into an ultrasonic extraction kettle to be ablated to obtain ablated mucus; adjusting the pH value of the ablated mucus to 5-8, and adding ethanol to make the ethanol to reach 10-40%; performing primary centrifugation, and collecting supernatant of the primary centrifugation; adjusting the pH value of the supernatant of the primary centrifugation to 2-4,and adding ethanol to make the ethanol to reach 40-70%; performing secondary centrifugation, collecting precipitate, and rinsing the precipitate with acidic ethanol; and redissolving the precipitate with buffer solution, and performing freeze-drying to obtain the white cotton mucin.
