Abstract

The invention relates to the field of biological detection, in particular to a high-throughput rapid detection method for perfluoro and polyfluoro compounds in serum. The method comprises the following steps: (1) serum pretreatment: adding lauryl sodium sulfate into sampled serum for pretreatment; (2) ion pair extraction: taking the pretreated serum to a 96-well plate for methyl tert-butyl ether centrifugal extraction; (3) ultrasonic extraction: freeze-drying the residual solution after extraction in the step (2), and then adding acetonitrile for ultrasonic extraction; (4) high-flux purification: adding the methyl tert-butyl ether extracting solution obtained in the step (2) into a PRiME HLB 96 pore plate, leaching by using the acetonitrile extracting solution obtained in the step (3), blowing methanol to a constant volume by using nitrogen, and centrifuging; and (5) transferring the supernatant obtained after centrifugation in the step (4) to a new 96-well plate for chromatography-mass spectrometry detection. The method has the advantages of high flux, low cost, rapidness and the like, and can be efficiently used for detecting the PFASs in environmental monitoring.

Description

technical field [0001] The invention relates to the field of biological detection, in particular to a high-throughput rapid detection method for perfluorinated and polyfluorinated compounds in serum. Background technique [0002] Perfluorinated and polyfluorinated compounds (PFASs) are linear or branched organic compounds in which the hydrogen atoms on the carbon chain are partially or completely replaced by fluorine atoms. Due to their excellent chemical stability and surface activity, they are often used as additives widely. Used in fire extinguishing agents, lubricants, coatings, toiletries, photosensitive material surface treatment agents, textile and leather finishing agents and other fields. Globally, PFASs pollution is very common, and studies have shown that they are widely detected in environmental media (atmosphere, water and soil), plants, animals and human samples. Toxicology studies have found that PFASs are bioaccumulative and potentially biotoxic, which may c...
