Embodiment 1

[0075] The preparation and use of the kit

Embodiment approach 1

[0076] Embodiment 1: The kit consists of magnetic microspheres coated with AMH antibody on the surface and AMH antibody labeled with alkaline phosphatase. The AMH antibody was labeled with alkaline phosphatase, and AMPPD was used as the luminescent substrate.

Embodiment approach 2

[0077] Embodiment 2: The kit consists of magnetic microspheres coated with AMH antibody and acridinium ester-labeled AMH antibody. The AMH antibody was labeled with acridinium ester, and hydrogen peroxide and sodium hydroxide were used as luminescent substrates.

Abstract

The invention relates to the field of antigen detection, in particular to a mullerian hormone determination kit for marking a luminescent indicator based on a reduction method. The kit comprises a solid-phase support and an AMH (Anti-mullerian hormone) capturing probe, wherein the solid-phase support is coated with an AMH first antibody; and the AMH capturing probe is an AMH second antibody markedwith a chemiluminescence signal strength indicator, wherein a method for marking the chemiluminescence signal strength indicator with the AMH second antibody comprises the following steps: cutting off a disulfide bond of the AMH second antibody with a reducing agent to make sulfhydryl group exposed, and then marking the chemiluminescence signal strength indicator on the AMH second antibody through a functional group capable of being connected with the sulfhydryl group. The kit, when used for detecting AMH, has the advantages of good stability, high sensitivity and wide linear range.

Description

technical field [0001] The invention relates to the field of antigen detection, in particular to a Mullerian hormone assay kit based on a reduction method for labeling a luminescent indicator. Background technique [0002] Anti-Mullerian hormone (AMH) is a dimeric glycoprotein belonging to the transforming growth factor β (TGFβ) family. All members of this superfamily are involved in the regulation of tissue growth and differentiation. Prior to secretion, the hormone undergoes glycosylation and dimerization to generate two identical disulfide-bonded 70 kDa subunits of an approximately 140 kDa precursor. Each monomer contains a large N-terminal proregion and a much smaller C-terminal maturation domain. In contrast to other TGFβ family members, AMH is thought to require the N-terminal domain to enhance the activity of the C-terminal domain for full biological activity. A portion of AMH is then cleaved at a site between the proregion and the mature domain during cytoplasmic ...
