[0035] The preparation of embodiment 1 Aspergillus aculeatus P93 microbial agent

[0036] 1. Isolation and purification of Aspergillus aculeatus P93

[0037]A bacterial strain P93 that can efficiently dissolve insoluble phosphorus was isolated and purified from farmland soil (calcareous soil) in Shijiazhuang City, Hebei Province.

[0038] According to the mycological characteristics of the strain P93 and the results of ITS rRNA gene sequencing, the strain P93 was identified as Aspergillus aculeatus.

[0039] 2. Cultivation of Aspergillus aculeatus P93

[0040] (1) Composition of medium (kg / t fermentation liquid)

[0041] Soluble starch 10kg, soybean cake powder 10kg, corn flour 5kg, glucose 1kg, sucrose 10kg, yeast powder 5kg, (NH 4 ) 2 SO 4 3kg,K 2 HPO 4 0.2kg, NaCl 2.5kg, MgSO 4 ·7H 2 O 0.1kg, CaCO 3 0.5kg, FeSO 4 0.001kg, boric acid (1%) 1L, pH7.0, prepared with water.

Embodiment 2

[0048] Embodiment 2 Bacterial strain P93 solid plate test phosphorus dissolving effect

[0049] In order to clarify the ability of Aspergillus aculeatus P93 to dissolve insoluble phosphorus, tricalcium phosphate Ca was used in this example 3 (PO 4 ) 2 As a phosphorus source, cultivate at 28°C for 5 days, and measure the available phosphorus on the PDA plate. Penicillium bayley ATCC 20851 and Penicillium oxalicum P8 (CGMCC 2272) were used as control strains in the test, and a control test without inoculation was set up.

[0050] The results show that P93 fungus has the strongest ability to dissolve tricalcium phosphate, and the dissolved phosphorus is as high as 48.16mg phosphorus / plate, which is 128.8% higher than the dissolved phosphorus (21.05mg phosphorus / plate) of bacterial strain ATCC 20851 (table 1); the dissolved phosphorus of bacterial strain P93 Slightly lower than Penicillium oxalicum P8, the amount of dissolved phosphorus is 48.76mg phosphorus / plate. Colonies fo...

Embodiment 3

[0054] The effect of embodiment 3 phosphate-dissolving bacterial strain P93 dissolving insoluble phosphorus in liquid medium

[0055] In liquid culture medium at 20°C, 17rpm, after 5 days of cultivation, the dissolution effect of phosphate-dissolving fungus P93 on tricalcium phosphate, aluminum phosphate, iron phosphate, zinc phosphate and phosphate rock powder was measured. The results showed that the Aspergillus aculeatus P93 strain released the highest available phosphorus from tricalcium phosphate, reaching 934.89 μg phosphorus / mL, accounting for 94.2% of the total phosphorus added; the strain P93 had a strong ability to dissolve aluminum phosphate, and the available phosphorus was 494.73 μg phosphorus / mL, respectively accounted for 39.0% of the total phosphorus added; strain P93 has a strong ability to dissolve iron phosphate, and the released available phosphorus also reached 344.6 μg phosphorus / mL, accounting for 33.6% of the added insoluble phosphorus; strain P93 dissol...

Abstract

The invention discloses aspergillus aculeatus P93, a biofertilizer preparation prepared from same and application thereof. The aspergillus aculeatus P93 has the preservation number of CGMCC NO.14150.Microbial inoculum prepared from the aspergillus aculeatus P93 has obvious effects of promotion of crop growth, yield increasing and activation of soil phosphorus. and the microbial inoculum P93 can dissolve multiple insoluble phosphorus, ground phosphate rock, iron phosphate, zinc phosphate and tricalcium phosphate are efficiently dissolved in a liquid culture medium, and dissolution rate reaches12.1-94.2%; under soil conditions, five insoluble phosphorus sources are used, then the microbial inoculum P93 is inoculated, and soil available phosphorus is improved by 257.8-1004.6%; the microbialinoculum P93 is inoculated for promoting phosphorus absorption of corns, and total phosphorus of a corn plant is increased by 3.4-136.4%; and phosphorus dissolving capability is higher than that of phosphorus dissolving penicillium bilaiae (ATCC 20851) which is reported at present and internationally recognized. The microbial inoculum disclosed by the invention has significance on conservation ofphosphorus resources in agricultural production in China, the sustainable yield increase of cereals and protection of farmland ecological environment.

Description

technical field [0001] The invention relates to the field of microbial preparations, in particular to Aspergillus aculeatus P93 and biological fertilizer preparations and applications prepared therefrom. Background technique [0002] Phosphorus is the second most important element for plant growth after nitrogen, but its availability is very low due to soil fixation, making it difficult to meet the needs of crop growth (Wild, 1988). Studies have shown that more than 90% of the phosphorus fertilizer applied to the soil is converted into insoluble phosphorus, acidic soil is converted into iron phosphate and aluminum phosphate, and calcareous soil is converted into calcium phosphate (Hagin and Hadas, 1962; Nagelschmidt G, et al. 1994). In order to increase crop yield, the amount of phosphate fertilizer has increased sharply, and phosphate rock resources are facing a crisis of depletion. It is estimated that the world's phosphate rock can be used for 60-90 years (Runge-Metzger ...
