Abstract

The invention discloses a separation, enrichment and detection method for a camptothecin-containing plant hydroponic root system allelochemical. The method comprises the following steps: firstly taking silica gel as a filler, filling a solid-phase extraction column, extracting and adsorbing root system metabolites for plant hydroponic solution, then taking out the filled silica gel, loading to an upper layer of a silica gel chromatography column, carrying out column chromatography isolation, then eluting in different combination ratios by taking petroleum ether, ethyl acetate, ethanol and chloroform as eluents, collecting eluate according to the composition of the eluate, carrying out rotary evaporation and concentration, and analyzing a separated component by utilizing a GC-MS technology. The method disclosed by the invention has the advantages that separation, enrichment and detection on a large number of allelochemicals in two kinds of plant hydroponic root secretions are realized; 25 allelochemicals are separated from camptotheca acuminate hydroponic root secretions and ophiorrhiza japonica hydroponic root secretions in total; and the method disclosed by the invention has very important technical guiding effect on ascertaining of more allelochemicals in a hydroponic system, substances with autotoxicity in the allelochemicals can be eliminated in time based on the method disclosed by the invention, plant growth cannot be inhibited, and vigorous growth of plants is promoted, so that the method disclosed by the invention has important significance on optimization of the plant hydroponic system.
