Embodiment 1

[0039] Using Pyromorpha indica to reduce heavy metal lead residues in hydroponic tobacco includes the following steps:

[0040] Activation of strains: Inoculate the Pyromorpha indica strains stored in the refrigerator on Kafer medium (KM) plates, and culture them in a 28°C incubator in the dark for 7 days. Use a hole punch with a diameter of 5 mm to punch holes at the edge of the activated Pyrocytosporum indica colony.

[0041] Bacteria culture: prepare liquid Kafer medium and liquid Kafer medium containing nanomaterial ZnO (30nm) 0.01g / L (that is, replace ZnSO4 with nanomaterial ZnO with a particle size of 30nm), and pack in 250mL triangular conical flasks , 150mL culture medium in each bottle, sterilized for later use. Under sterile conditions, add the mycelia blocks of Pyromorpha indica into the Erlenmeyer flask containing the above medium, 6 pieces of mycelium blocks per bottle; place in a constant temperature shaker at 120rpm, 28°C and avoid light for 5- 7 days.

Embodiment 2

[0060] In Example 1, the mycelia block of Pyriformis indica was changed into Pyromorpha indica spore liquid, inoculated in the liquid Kafer medium containing 10mg / LZnO, and cultured in a constant temperature shaker at 120rpm and 25°C in the dark for 7 days, and the rest Identical to Example 1, the final accumulation of lead in hydroponic, potted and field tobacco leaves was reduced by 60.1%, 38.6%, and 32.1%, respectively.

Embodiment 3

[0062] In Example 1, the concentration of Pyromorpha indica mycelia liquid was changed to 2g / L, and the rest were equal to Example 1. Finally, the accumulation of lead in hydroponics, potted plants and field tobacco leaves decreased by 61.2%, 31.1%, and 29.9% respectively.

Abstract

The invention provides a method for reducing lead content of tobacco leaves. According to the method, parasitic piriformospora indica at the root of tobacco is utilized to reduce heavy metal lead residues in tobacco. The method comprises the following specific steps: (1) by use of utilizing nano material ZnO to replace ZnSO4.7H2O in an original Kafer culture medium, adding 10mg of nano material ZnO into per litre of the culture medium; (2) inoculating the activated piriformospora indica into the liquid Kafer culture medium obtained in the step (1) for culturing, then collecting mycelia, preparing a piriformospora indica mycelium solution, then pouring the piriformospora indica mycelium solution to the root of a tobacco seedling, and performing root-irrigation treatment so as to colonize the piriformospora indica at the root of the tobacco seedling; (3) after performing piriformospora indica colonization, transplanting the tobacco seedling into a large field, and then performing routine water and fertilizer management so as to enable the tobacco to grow normally. The method can be used for improving the tobacco growth condition, reducing the transfer of the heavy metal lead from a root system to the overground part of the tobacco and reducing the heavy metal lead residues, and is easy to master, relatively low in cost, simple and practical, environment-friendly, and incapable of causing secondary pollution.

Description

technical field [0001] The invention relates to the technical field of the utilization of microbial resources and the quality and safety of agricultural products, in particular to a method for reducing the lead content in tobacco leaves. Background technique [0002] With the extensive use of chemical fertilizers and pesticides and the rapid development of industry, environmental pollution has become increasingly serious. Lead is a toxic heavy metal and one of the main environmental pollutants. Tobacco is one of the important economic crops in my country. The planting area, flue-cured tobacco output and production volume of China rank first in the world. In recent years, there have been reports of quality and safety issues such as excessive heavy metals in tobacco, among which cadmium, chromium, and lead have a more serious impact on tobacco growth and quality. Lead in the soil can inhibit the growth of tobacco, change the morphology, physiology, biochemistry and structure...
