Problems solved by technology

[0013] The inventor’s previous research results and a small number of literature reports show that the extract of peony angelica powder or paeoniflorin can inhibit the activation of microglial cells caused by lipopolysaccharide or cerebral ischemia, but the mechanism of microglial cell activation under this condition is different from that of opioids. The mechanism of the induced microglial activation is obviously different. Therefore, whether paeoniflorin can also inhibit opioid-induced microglial activation and p-P38 up-regulation to exert its opioid analgesic enhancement and delay or reduce opioid tolerance effect is unknown

Embodiment 1

[0046] Example 1. After the analgesic effect of intrathecal administration of morphine disappears, the administration of paeoniflorin can reproduce the significant analgesic effect

[0047] a) Test animals:

[0048] Healthy adult male SD (Sprague-Dawley) rats, clean grade, weighing 180-220g. The experimental animals were raised in an independent environment with 12h-12h alternation of day and night, the room temperature was maintained at 24±2°C, water and food were free to drink, and the experiment was carried out after 1 week of adaptation to the environment. All handling of animals followed the requirements of the Ethics Committee of the International Association for the Study of Pain.

[0049] b) Test drugs and reagents:

[0050] The drug paeoniflorin, with a purity of 98%, is commercially available. Morphine hydrochloride was purchased from Shenyang No. 1 Pharmaceutical Factory.

Embodiment 2

[0059] Example 2. Intrathecal administration of compound preparations of paeoniflorin and morphine can enhance the acute analgesic effect of morphine

[0060] a) Test animals: healthy adult male SD rats, clean grade, with a body weight of 180-220 g.

[0061] b) Test drug: obtained by corresponding dilution of the compound preparation of paeoniflorin and morphine prepared in Example 12.

[0062] c) Test method: Intrathecal injection of rats: Press both sides of the lumbosacral area of the rat to fix it, insert the needle from the L5-L6 spinous process, and the sudden lateral m...

Embodiment 3

[0067] Example 3. Systemic administration of paeoniflorin can enhance the acute analgesic effect of systemic administration of morphine

[0068] a) Experimental animals: healthy adult female ICR mice, clean grade, weighing 18-22 g.

Abstract

The invention discloses novel application of paeoniflorin in the field of analgesics, and in particular relates to application of paeoniflorin in preparation of a medicine for reinforcing the analgesic effect of opioid medicines, and retarding or reducing the opioid tolerance. The application of paeoniflorin matched with opioid medicines or compound preparation prepared from paeoniflorin and opioid medicines can be used for prolonging the analgesic time of opioid medicines and strengthening the analgesic effect of opioid medicines, and can retard or reduce the opioid tolerance phenomenon in the treatment process of opioid medicines. The effect can be realized by using p-P38 of paeoniflorin in inhibiting opioid medicine induced over activation of spinal microglial cells and inhibiting opioid induced over up-regulation.

Description

technical field [0001] The invention belongs to the field of pharmacy, and relates to the new use of paeoniflorin in the field of analgesic drugs, in particular to the use in preparing drugs for enhancing the analgesic effect of opioid drugs and delaying or alleviating opioid tolerance. Background technique [0002] Pain is a complex physiological and psychological activity, including the pain sensation caused by noxious stimuli acting on the body, and the body's pain response to noxious stimuli. Pain can be used as a warning that the body is injured, causing a series of defensive protective reactions of the body, but some acute or long-term severe pain is an unbearable torture to the body. Pain is one of the most common clinical symptoms, and it is also an important problem that medical workers need to solve urgently. [0003] In terms of the course of the disease, pain can be divided into acute pain and chronic pain. Acute pain is pain that is new and of short duration, ...
