Problems solved by technology

However, existing studies have shown that some pathogenic bacteria targets have a high frequency of spontaneous mutations that are resistant to such agents, and pathogenic bacteria are prone to form drug-resistant populations under the high selection pressure of agents.

Embodiment 1

[0030] Example 1. Isolation and identification of biocontrol fungus HTC

[0031] 1. Strain isolation and identification

[0032] (1) Separation of strains

[0033] Walnut leaves collected from the Walnut Production Base of Lulaishan Forest Farm in Tai'an City, Shandong Province were rinsed with tap water, and then rinsed twice with sterile water, and the surface water was absorbed by sterile filter paper. Under aseptic conditions, cut the walnut leaves into 4×8mm tissue pieces, soak in 70% ethanol for 30sec, rinse 3 times with sterile water, and transfer to 1‰ mercury disinfectant (1g mercury, 2.5ml concentrated hydrochloric acid, 1000ml water ) Soak for 3 minutes, rinse with sterile water 3 times, and absorb the surface water with sterile filter paper. Place the sterilized walnut leaf tissue block evenly on the prepared PDA medium plate at 5 points and place it at 25℃ for constant temperature incubation. After 3 days, observe it daily. When the hyphae grow from the edge of the lea...

[0053] Example 2: Application of biocontrol fungus HTC in preventing and controlling pepper blight

[0054] 1. Preparation of HTC biocontrol preparations

[0055] (1) Preparation of seed liquid:

[0056] The strain of Chaetomium aureum HTC CGMCC No.6173 isolated and preserved in Example 1 stored on the slope of the test tube at 4°C was transferred to a fresh PDA medium plate for activation, and cultured at 25°C for 5 days After growing fresh hyphae, they were transferred to a large number of fresh PDA plates and cultured at 25°C for 10 days. After a large number of ascospores are produced, they are scraped off under aseptic conditions and prepared with sterile water to a concentration of 1×10 8 The spore suspension of cfu / ml is the HTC seed solution.

[0057] (2) Preparation of biocontrol agents:

[0058] The HTC seed liquid was added to the PDB medium at a volume ratio of 1:100, and cultured with shaking at 160 rpm at 25°C for 12 days; filtered out the mycelium in the fermentation pr...

Abstract

The invention discloses a biocontrol strain HTC for preventing pepper phytophthora blight and application thereof, and provides a Chaetomium aureum strain HTC. The preservation number is CGMCC (China General Microbiological Culture Collection Center) No. 6173. The invention further provides a method for preparing an HTC biocontrol agent for preventing the pepper phytophthora blight. The method comprises steps as follows: the Chaetomium aureum strain HTC is fermented, so that a fermentation product is obtained, namely, the biocontrol agent is obtained. The experiment of the invention proves that in a greenhouse, the strain HTC biocontrol agent has a preventive effect on the pepper phytophthora blight to a certain extent, the effect can reach 72.19%, the strain height is increased by 12.27% and the live weight is increased by 32.28% in a seedling stage; and the live weight is increased by 31.11% and the yield of a pepper fruit is increased by 21.81% in reaping time.

Description

Technical field [0001] The invention relates to the field of biotechnology, in particular to a biocontrol strain HTC for preventing and curing pepper blight and its application. Background technique [0002] Pepper blight is an oomycete disease caused by Phytophthora capsici. Phytophthora belongs to the phylum Oomycota, Peronospora, Pythium, and Phytophthora. Phytophthora survives in the soil not in one form, but in different forms, namely mycelium, sporangia, chlamydospores and oospores. The disease can be spread through rainwater, irrigation water, and agricultural equipment. It is a very harmful soil-borne disease. With the large-scale planting of peppers in my country, pepper blight has also occurred on a large scale in open fields, protected areas and greenhouses. Because the disease has a short onset cycle and a fast epidemic rate, it often leads to the death of pepper plants in pieces and brings serious economic losses to the pepper industry. The general diseased plant ...
