Abstract

The invention discloses application of prosaposin (psap) serving as an effect marker in detection of the male reproductive toxicity of polychlorinated biphenyl, and relates to psap. The psap is multifunctional glycoprotein, has the molecular weight of 70kD, and exists in some biological fluids. The method for screening the psap comprises the following steps of: exposing a small mouse by using different quantities of polychlorinated biphenyl, and acquiring a sample; screening difference expression genes of the sample; performing function accumulation on the screened difference expression genes to obtain a difference expression gene relevant to a reproductive function; detecting and analyzing the change of the obtained difference expression gene on an mRNA transcription level and a protein expression level by using a real-time fluorescent quantitative polymerase chain reaction (PCR) technology and an immunoblotting method along with the increase of a polychlorinated biphenyl (PCB) exposed amount and the exposing time; detecting and analyzing sperm parameters; and counting relevant coefficients among the effect marker, an in-vivo polychlorinated biphenyl level and a semen index through Spearman correlation analysis.
