Embodiment 1

[0141] Embodiment 1: the synthesis of iron ion two-photon fluorescent probe

[0142] 1 mmol tetrabutylammonium bisulfate, 22 mmol 4-diethylaminobenzaldehyde and 10 mmol 2-amino-4,6-dimethylpyrimidine were refluxed in 50 mL 5M NaOH solution for 2 hours. The orange product was obtained by rotary evaporation, and the pure material was obtained by column chromatography (ethyl acetate:petroleum ether=1:1).

Embodiment 2

[0143] Example 2: Synthesis of α-synuclein two-photon fluorescent probe

[0144] 56 mmol ruthenium trichloride, 112 mmol o-phenanthroline and 3.7 mmol lithium chloride were refluxed in 25 mL DMF for 8 hours. Acetone was then added to the reaction mixture, which was cooled overnight at 4°C, and the product Ru(phen) 2 Cl 2 Used without further purification. 12mmol Ru(phen) 2 Cl 2 And 10mmol dipyrido[3,2-A:2'.3'-C]phenazine was refluxed in 1:1 methanol aqueous solution for 4 hours. The apparatus was cooled to room temperature, ammonium hexafluorophosphate was added to precipitate the product from solution and filtered to give crude product. Purification by column chromatography (dichloromethane:acetonitrile=4:1) and recrystallization (ethanol:water=90:10) gave red-orange crystals.

Embodiment 3

[0145] Embodiment 3: the synthesis of multifunctional biological nanocomposite material

[0146] In PBS buffer solution with pH=7.4, 20 μg 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride was added to 5 mL of 10 μg / mL copper sulfide nanospheres and stirred for 2 hours, then 20 μg of N-hydroxysuccinimide was added and stirred for half an hour, after which 100 μL of 50 μg / mL iron ion two-photon fluorescent probe prepared in Example 1 of the present invention was added to the mixture overnight under stirring. The product was collected by centrifugation and purified by washing several times with deionized water, then resuspended in 5 mL of PBS buffer solution, pH=7.4. Finally, 100 μL of 50 μg / mL α-synuclein two-photon fluorescent probe prepared in Example 2 of the present invention was added to the solution and stirred overnight at room temperature. The resulting product was collected by centrifugation, purified by washing several times with deionized water, and then r...

Abstract

The invention discloses a multifunctional biological nanocomposite material and a synthesis method thereof. The nanocomposite material is formed by compounding an iron ion two-photon fluorescent probe, an alpha-synuclein two-photon fluorescent probe, porous hollow copper sulfide nanoparticles, a DNA nanodoor and an alpha-synuclein aggregate therapeutic drug. The two two-photon fluorescent probes are short in synthesis steps and simple to operate, and have high selectivity and high sensitivity; the copper sulfide nanoparticles are synthesized by a one-pot method, are uniform in size distribution and large in specific surface area, and have good drug loading capacity. The invention also discloses an application of the multifunctional biological nanocomposite material in in-vitro and in-vivotwo-photon fluorescence lifetime detection and imaging of iron ions and alpha-synuclein aggregates. The interaction relationship between iron ions and alpha-synuclein related to Parkinson's disease isanalyzed for the first time, and a diagnosis and treatment integrated platform of chemotherapy and photothermal therapy is constructed, and the multifunctional biological nanocomposite material is ofgreat significance to research on treatment and diagnosis of Parkinson's disease.

Description

technical field [0001] The invention belongs to the technical field of nanometer material analysis and detection, and relates to a biological nanocomposite material and its synthesis method and application. Background technique [0002] Parkinson's disease is a common neurodegenerative disease, first proposed by James Parkinson in 1817, and it is the second most common neurodegenerative disease in humans after Alzheimer's disease. Its etiology and pathogenesis have not yet been clarified, and the pathology mainly includes the degeneration and death of dopamine neurons in the substantia nigra of the midbrain, the significant reduction of the striatum, and the appearance of Lewy bodies in the cytoplasm of neurons in the substantia nigra. α-synuclein is the main component of Lewy bodies. It is located at the presynaptic terminal of neurons. It is a protein composed of 140 amino acids. Its mutation, aggregation and excessive accumulation are related to a series of diseases inclu...
