The method comprises the steps that n-butyl alcohol, ethyl acetate, methanol and water are prepared into solvent systems of a flow phase and a fixed phase; the separation column of a counter-current chromatograph is fully filled with the fixed phase, and the flow phase is pumped to achieve two-phase balance; after sample feeding of a sample feeding valve, the flow phase is eluted; a target component is collected according to a detector's graph and combining HPLC detection; an oleuropein monomer compound is obtained after fractions containing the oleuropein are mixed and solvents are recovered. By adopting the method, the oleuropein compound having the purity 90% or above can be obtained.
