Abstract

The invention discloses an orchid seed preserving and seedling raising method. The method comprises the steps: harvesting orchid capsules developed basically ripe and having no cracking, disinfecting, transferring seeds in the capsules into a sterilized centrifugal tube under an aseptic condition, covering with a centrifugal tube cover and sealing with a breathable membrane, and preserving under a condition of the temperature of 4-6 DEG C; and when seedling raising is needed, taking out the preserved seeds, placing to the centrifuge tube at room temperature to reach room temperature, making the seeds into a seed suspension with sterile water, taking the seed suspension and then evenly inoculating a one-step seedling formation culture medium with the seed suspension, and culturing into seedlings by one time. After the orchid seeds are stored for 5 years under the condition of the temperature of 4-6 DEG C, the orchid seeds still maintain a relatively high germination rate (75.6%); moreover, required facility conditions are simple, cheap, pollution-free and huge in preserving capacity; the seeds preserved by the method have no need for surface sterilization treatment, are subjected to aseptic sowing by using a non symbiotic germination technology, and can be cultured into seedlings at one time.
