Embodiment Construction

[0018] It is believed that there is a need for a disposable pH sensor compatible with plastic bag type, ready to use, disposable bioreactors. Many pH sensors based on glass electrodes require protection of the sensor's reactive surface or membrane from physical or environmental damage. This function is usually provided by a disposable "shield" or cap that covers the sensing end of the sensor.

[0019] Embodiments of the present invention generally include a pH sensor mounted to the wall of a single-purpose bioreactor / mixer, wherein the bioreactor / mixer has a bacteria (gamma ray irradiation), mechanical structures that allow protective solutions to be stored around the pH sensing and reference elements during storage and transport. The mechanism also allows the storage cavity holding the protective solution to be opened to expose the detection and reference elements prior to manipulation.

Abstract

The invention provides a pH value detection biological reaction system. The system comprises a bioreactor vessel (51) having a plastic wall (50) and a pH sensor (40, 60, 80, 100, 120) attached to said plastic wall (50). The pH sensor (40, 60, 80, 100, 120) includes a sensor body (82) having a flange (76) sealingly attached to the plastic wall (50). The sensor main body (82) has a reference electrolyte (142) and a first detection element (152) arranged in the reference electrolyte (142). The first detection element (152) is configured to contact both the reference electrolyte (142) and the sample solution (52) within the bioreactor vessel. A second detection element (62, 92, 148) is positionable inside the bioreactor vessel (51). The pH sensor (40, 60, 80, 100, 120) has multiple configurations, including a protective configuration in which the at least one detection element is isolated from the interior of the bioreactor container (51), and the at least one detection element is fluid ground connected to the internal working configuration of the bioreactor vessel (51).

Description

Background technique [0001] pH determination is one of the most common process chemical measurements performed today. The pH value is expressed as a measure of the relative amounts of hydrogen ions and hydroxide ions in an aqueous solution. In fermentation and cell culture, one of the toughest procedural challenges is maintaining ideal pH levels. Fermentation processes use living organisms, such as yeast, bacterial, or fungal strains, to produce active ingredients. The fermentation process usually has a relatively short duration (2-7 days). Cell culture is the process by which mammalian cells are grown to produce active ingredients. The cell culture process usually takes longer (2-8 weeks). [0002] An important challenge for pH determination in the field of fermentation and cell culture involves the cleaning process of the fermenter or bioreactor. In particular, fermenters or bioreactors must be sterilized before commencing the corresponding procedures to ensure avoiding...
