[0005] 2) The detection limit is too narrow At this stage, the monitoring and measuring instruments used by most scientific research institutions and environmental monitoring stations in China are DXY-2 fluorescence detectors developed by the Nanjing Soil Institute of the Chinese Academy of Sciences. This instrument was developed in the 1990s and has low sensitivity. Good, limited ability to detect the toxic effects of low concentrations of pollutants on luminescent bacteria

Embodiment

[0050] Determination of the Toxicity of Acetone to Photobacteria

[0051]Due to the volatility of acetone, studies have shown that the biotoxicity data for this substance are not stable. Using this method, use 3% NaCl to configure 0.034, 0.061, 0.872, 0.109, 0.195, 0.347 mol / L acetone solutions and add them to the measurement system according to the above method. Each concentration gradient is set up in 3 groups in parallel, and each group is repeated 3 times.

Abstract

The invention belongs to the technical field of detection of environmental pollution, and discloses a method for detecting the toxicity of luminescent bacteria. The method comprises the following steps of: 1, activating strains, namely dissolving freeze dried powder of photobacterium phosphoreum, inoculating on a slant and generating the strains for 3 times for later use; 2, culturing the strains, namely inoculating the generated strains to a liquid nutrient medium by using an inoculating loop and culturing for 12 hours at the temperature of 20 DEG C and at 180rpm; 3, balancing bacteria liquid, namely adding an appropriate amount of the bacteria liquid into 20 milliliters of NaC1 solution at the concentration of 3 percent, controlling light value measurement ranges to be between 0.5 and 5million, and agitating for 40 minutes at the temperature of 20 DEG C; 4, adding samples and detecting luminous intensity, namely adding the bacteria liquid which is agitated for 40 minutes into 200 to 800 microliters of NaC1 blank solution at the concentration of 3 percent and 800 microliters of NaC1 polluted solution at the concentration of 3 percent by using a continuous sample injector, immediately and uniformly mixing for 2 minutes on a vortex mixer, standing for 15 minutes at the temperature of 20 DEG C, and placing a sample in a fluorescence detector to detect the luminous intensity; and 5, calculating the suppression ratio of the sample to the luminous intensity by a double alternate comparison method. The method has the characteristics of convenience and high precision.

Description

technical field [0001] The invention belongs to the technical field of environmental pollution detection, and in particular relates to a method for measuring the toxicity of luminescent bacteria. Background technique [0002] With the improvement of people's requirements for the quality of life and the quality of the environment, people pay more and more attention to the pollutants in the environment. Most of the pollutants in the environment have the characteristics of low concentration and compound type. Therefore, the low sensitivity of previous instruments has caused many low-concentration pollutants to be ignored, but some of them contain more toxic pollutants. Taking POPs (Persistent Organic Pollutants) as an example, it is a class of organic pollutants with long-term persistence, bioaccumulation, semi-volatile and high toxicity. However, it has the characteristics of low water solubility and high fat solubility. If the sensitivity of the monitoring instrument is not...
