Abstract

The invention discloses a method for extracting melanin from cordyceps militeris fungus fermentation broth. The method includes the following steps: mycelium is separated from the cordyceps militeris fungus fermentation broth, and the fermentation broth is acidized to obtain melanin crude extract after the sediment is collected. The melanin crude extract is hydrolyzed by hydrochloric acid to eliminate protein impurities, and is repeatedly scrubbed by organic solvent and dissolved by alkaline solution, and is acidized to form sediment for purification, and then the chloride ions are eliminatedby deionized water through repeated scrubbing, finally the cordyceps militeris melanin end product is obtained after drying. The melanin prepared by the invention has similar properties with melanin CAS: 8049-97-6 sold by Sigma Corporation. Both two types of melanin can dissolve in alkaline solution, cannot dissolve in organic solvents, such as water, ethanol, acetone, chloroform and hexane, completely become sediment when pH value is less than 3 and can fade under the effect of strong oxidizer; the ultraviolet visible light area scan map includes a curvature with reducing absorbance along with the increasing of wavelength, and strong absorbing bands are respectively near 340cm-1 and 1602cm-1 in the infrared spectrum.
