[0063] 1. Methods and materials

[0064] 1. Clinical samples

[0065] Clinical samples were provided by patients with written informed consent, and were approved by the Ethics Committee of Yueyang Hospital ( Figure 25 ). All patients underwent surgical treatment in surgery from April 2019 to February 2020. DU samples were collected from the margins of unhealed wounds during surgical debridement, and normal skin samples were collected from excess skin collected during cosmetic surgery. Immediately after the tissue was cut, it was fixed with 70% formalin and embedded in paraffin. The clinical characteristics of the patients are shown in Table 1.

Abstract

The invention relates to application of an eIF3I-PDL1-IRS4 axis in preparation of a medicine for treating refractory ulcer. It is found that: when PDL1 is used for treating wounds, tissue formation, inflammation fading and reduction in vascularity can be induced; epithelization is accelerated; inflammation is relieved; healing of diabetic ulcer (DU) wounds is helped; in a DU wound surface, the expression of IRS4 is increased; the expression of the IRS4 of HaCaT cells is knocked down; the proliferation and migration of the cells are increased; when the eIF3I is over-expressed in HaCaT cells, the proliferation and migration of the cells are remarkably increased; and inflammatory infiltration is relieved. The invention prompts that the targeted eIF3I-PDL1-IRS4 axis can be used as a novel treatment method for treating refractory ulcer, chronic inflammation after injury, infection and other types of wounds.

Description

technical field [0001] The invention relates to the technical field of biomedicine, in particular to the application of the eIF3I-PDL1-IRS4 axis in the preparation of refractory ulcer medicine. Background technique [0002] The skin is the organ that the human body has the most contact with the environment, and is directly exposed to the surrounding environment. It has functions such as sensory and immune protection, and is also vulnerable to injury and infection. Wound healing is impaired in diabetes, vascular disease, and elderly patients. As the diabetic population grows, the impact of diabetic ulcers (DUs) on healthcare costs is becoming increasingly significant. Diabetic ulcer is a complication of diabetes, which occurs mostly on the feet of patients, so it is also called diabetic foot. Patients with long-term diabetes often have vascular lesions and complications of peripheral neuritis. The feeling of the toes of such patients is often lost due to neuritis, so they ...
