The preparation method comprises the following steps: dissolving a polysaccharide substancewith an amino group and protein in an acidic solution to obtain a precursor solution, dissolving spermidine and micromolecular dialdehyde or a high polymer material of which two ends are modified byaldehyde groups into absolute ethyl alcohol to obtain a spermidine cross-linking agent; employing a 3D biological printing method, printing a precursor solution in a culture dish, atomizing the spermidine cross-linking agent, controlling the hardness of each layer by controlling the concentration of the spermidine cross-linking agent of each layer, and finally obtaining the bionic anti-rejection artificial skin with the hardness changing in a gradient mode. According to the invention, the problem of a rejection reaction caused by animal-derived natural polymer artificial skin is solved by utilizing natural anti-rejection biological small molecular spermidine; artificial skin with strength gradient and microstructure is prepared by using a 3D printing method, and the problem that an existing skin substitute is not matched with the biomechanical property of human skin is solved.
