Embodiment 1

[0028] Add 2.0 g of Corydalis extract to 250ml reaction bottle (the content of tertiary amine base is 0.5% in terms of tetrahydropalmatine, figure 1 ), add methanol 100 ml, stir for 0.5 h, add K 2 CO 3 3.0 g, stirred for 10 min, added dropwise with 0.12 g NaBH 4 Add 2.4 ml of 5% sodium hydroxide solution in 2 min, stir for 2 h, filter, concentrate the filtrate to remove methanol, add 30 ml of water, adjust the pH to 7-8 with 1 N hydrochloric acid, add 30 ml of dichloromethane to extract three times , combined, washed once with saturated sodium chloride, dried over anhydrous sodium sulfate, concentrated to obtain 0.05 g of a yellow solid, analyzed by HPLC, wherein the tertiary amine alkali content was 50.2% in terms of tetrahydropalmatine, see figure 2 , the comparative spectrum of the sample solution before and after the reaction is as follows image 3 As shown, the peaks of two main quaternary ammonium bases in the extract—palmatine and dehydrocorydalline disappeared in ...

Embodiment 2

[0030] Add 2.0 g of Corydalis extract to a 250ml reaction bottle, add 100 ml of ethanol, stir for 0.5 h, add 3.0 g of K2CO3, stir for 10 min, add 2.4 ml of 5% sodium hydroxide solution of 0.12 g NaBH4 dropwise, add dropwise for 2 min After completion, stir for 2 h, filter, concentrate the filtrate to remove ethanol, add 30 ml of water, adjust the pH to 7-8 with 1 N hydrochloric acid, add 30 ml of ethyl acetate to extract three times, combine, wash with saturated sodium chloride once, anhydrous sulfuric acid Drying over sodium and concentration gave 0.04 g of a yellow solid, which was analyzed by HPLC, wherein the content of the tertiary amine base was 55.6% in terms of tetrahydropalmatine.

Embodiment 3

[0032] Add 2.0 g of Corydalis extract to a 250ml reaction bottle, add 50 ml of ethanol, stir for 0.5 h, add 3.0 g of K2CO3, stir for 10 min, add 0.1 g of NaBH4 in 2.0 ml of 5% sodium hydroxide solution dropwise, add dropwise for 2 min After completion, stir for 2 h, filter, concentrate the filtrate to remove ethanol, add 30 ml of water, adjust the pH to 7-8 with 1 N sulfuric acid, add 30 ml of ethyl acetate to extract three times, combine, wash with saturated sodium chloride once, anhydrous sulfuric acid Drying over sodium and concentration gave 0.03 g of a yellow solid, which was analyzed by HPLC, wherein the content of the tertiary amine base was 60.5% based on tetrahydropalmatine.

Abstract

The present invention provides a preparation method of tertiary amine alkali components in rhizoma corydalis. The preparation method of the provided components is as follows: berberine quaternary ammonium alkali in the rhizoma corydalis extract is reduced into protoberberine tertiary amine alkali, wherein two quaternary ammonium alkalis with highest content in the rhizoma corydalis, namely palmatine and dehydrocorydaline, are respectively reduced into tetrahydropalmatine and corydaline, pH of a reaction supernatant is adjusted to be neutral, dry-steaming is conducted, and drying is conducted after desalting to obtain the tertiary amine alkali components, wherein a total alkali content is 50% or more. The preparation method is simple in process, high in efficiency, high in recovery rate andlow in production cost, and provides a good technical scheme for developing safe, effective and controllable rhizoma corydalis alkali component preparations.

Description

technical field [0001] The invention relates to the field of drug synthesis, in particular to a preparation method of tertiary amine base components in Corydalis Corydalis. Background technique [0002] Corydalis yanhusuo (Yuanhu) is the tuber of Corydalis yanhusuo W. T. Wang, a plant of the poppy family. It has the effects of promoting blood circulation, removing blood stasis, regulating qi and relieving pain. Modern medicinal chemistry and pharmacology experiments show that the main active ingredients in Corydalis Corydalis are two types of alkaloids, tertiary amine and quaternary ammonium, of which the content of tertiary amine base is about 0.65%, and the content of quaternary ammonium base is about 0.3%. (He Kai, Gao Jianli, Zhao Guangshu. Research progress on chemical constituents, pharmacological effects and quality control of Corydalis Corydalis, Chinese Herbal Medicine, 2007, 38(12): 1909-1912). These two types of ingredients are not only different in structure, bu...
