Embodiment 1

[0025] WT mice were taken, and after intraperitoneal injection of 500mg / kg acetaminophen or 200mg / kg DEN, serum was collected at 0, 6, 12, 24, and 48 hours for blood biochemical detection of ALT, AST, and Western Blot and RT- The expression of OPTN was detected by PCR method. figure 1 The results showed that with the occurrence of liver injury, the expression of OPTN increased, and after the recovery of liver injury, the expression of OPTN returned to the normal level.

Embodiment 2

[0027] Take WT and Optn - / - For mice, after intraperitoneal injection of 500mg / kg acetaminophen, blood serum was collected at 0, 6, 12, 24, and 48 hours for blood biochemical detection of ALT and AST, and liver tissue was fixed with 10% formaldehyde solution for 7 days, and then paraffin was prepared Embedded tissue blocks. Paraffin sections with a thickness of 8 microns were prepared from the samples for H&E staining. figure 2 The results showed that OPTN deletion significantly aggravated the severity of acute liver injury.

Embodiment 3

[0029] The samples in Example 1 were taken, and Western Blot was used to detect the expression of autophagy-related proteins LC3 and p62. image 3 The results showed that autophagy was activated as liver injury occurred.

Abstract

The invention provides application of optineurin protein in preparing acute hepatic damage diagnosis and treatment products. Experiments prove that the OPTN (optineurin) protein is increased in protein in acute hepatic damage tissues and relieves polarization of macrophages through autophagy to release acute hepatic damage, and accordingly acute hepatic damage can be relieved by expressing the OPTN protein or applying bavachinin. Therefore, the OPTN protein can serve as a specific marker protein for diagnosis and drug therapy of acute hepatic damage, and through the agonist of the bavachinin and overexpression viruses, effectively treats acute hepatic damage to achieve more accurate and quicker diagnosis and treatment of acute hepatic damage. The optineurin protein provides a new therapeutic target and an effective new drug for treating acute hepatic damage.

Description

technical field [0001] The invention belongs to the field of biopharmaceuticals and relates to the application of an optineurin (OPTN) protein in the preparation of products for diagnosing and treating acute liver injury. Background technique [0002] Acute liver injury refers to the damage of liver cells caused by various etiologies in patients without chronic liver disease. There are many reasons for acute liver injury, mainly including viral infection, inappropriate medication, and excessive alcohol intake. Most patients can maintain the main function of the liver and run normally. Clinically, mild cases show elevated serum transaminases and bilirubin; severe cases may develop liver failure, coagulation dysfunction, and hepatic encephalopathy. Acute liver failure is a challenging medical emergency that requires aggressive treatment. [0003] Optineurin (OPTN) protein is encoded by the OPTN gene, and it is mainly used clinically to treat primary open-angle glaucoma (POAG...
