Problems solved by technology

[0006] In response to the above limitations, Jiang Xingyu's research group at the National Center for Nanoscience and Technology developed a polydopamine-based one-step printing method for cell patterning, which is suitable for a variety of substrates, such as gold surfaces, glass, silicon surfaces, polystyrene (PS) surface, etc., have the characteristics of easy operation and low cost, but also have the disadvantages of opaque patterns and long time-consuming

[0032] 1. Preparation of oligodopamine

[0033] Put the dopamine monomer molecule into a Tris-HCl buffer solution with a pH of 8.5 for 8 hours to obtain a substance with a black precipitate. The solution was centrifuged at 15000 rpm to obtain a yellow supernatant solution containing oligodopamine.

[0034] 2. Print oligodopamine patterns on the surface of the quartz plate modified with polyethylene glycol silane

[0035] (1) Oxidize the quartz plate

[0036] Quartz sheet (8*8*0.5mm 3 , Purchased from Donghai County Weida Quartz Products Co., Ltd.) Followed by ultrasonic cleaning in acetone and absolute ethanol for 10 minutes, then boiling in a mixture of concentrated sulfuric acid and hydrogen peroxide (volume ratio 3: 1) for 1 hour, cleaning with ultrapure water, and drying . The quartz piece was placed in a low-temperature plasma processor to oxidize for 6 minutes.

Abstract

The invention provides a novel microcontact printing system for cell patterning. The system comprises oligomeric dopamine formed by dopamine monomers through polymerization. The molecular weight of the oligomeric dopamine is between 1000 and 10000 D; and a structural formula is shown in the description. A substrate is firstly subjected to chemical modification treatment to obtain a bioadhesion-resistant inert substrate; then, a water solution of the oligomeric dopamine is prepared; a stamp is prepared; next, the stamp is soaked into the water solution of the oligomeric dopamine; and next, thestamp adsorbing the oligomeric dopamine is stamped onto the inert substrate, so that a pattern on the stamp is obtained from the surface of the substrate. The patterning substrate can also be used forcell patterning growth. The realization of the system is simple. The existing technology for cell patterning is simplified.

Description

Technical field [0001] The present invention relates to a novel micro-contact printing system for cell patterning, and more specifically, to a novel micro-contact printing system for cell patterning. Background technique [0002] Cell patterning is an in vitro cell culture technology. Specifically, a certain patterned substrate is prepared to induce the cell's own response mechanism to adhere to the designed area to form a pattern. This method is different from the direct cultivation of cells in a petri dish, which is commonly used in the laboratory, which usually forms a cell monolayer. [0003] Cell patterning technology has become an important tool for studying and controlling the biological behavior of cells. It is widely used to study the interactions between cells, cells, and substrates (including cell adhesion and apoptosis, cell polarization behavior, Cell migration rate, cell differentiation behavior, etc.); in vitro models can also be constructed for use in tissue engine...
