Problems solved by technology

[0008] This technology has only been studied abroad, and there is no related technology research in my country; in addition, this technology needs to prepare a large number of purified antigens for immunization experimental animals; relevant antiserum and other reagents need to be imported from abroad, and there is no domestic production unit, so the import cost is high

Embodiment Construction

[0038] The specific implementation of the present invention will be described in conjunction with the examples.

[0039] Such as figure 1 Shown, the preparation method of koi herpes virus antiserum:

[0040] (1) Preparation of immunized animals

[0041] Select healthy koi immunized animals, from farms with no history of koi herpes virus disease (KHVD), have not been vaccinated against koi herpes virus disease, weigh 150-350g, and have a body length of 15-25cm. Before inoculation, they should be cultured at room temperature (water temperature not exceeding 30°C) for no less than one week, and fed regularly in small amounts. Before inoculation, a small amount of blood was collected from the tail vein, and the serum was separated, used as negative serum, and frozen for future use. In order to prevent the death of koi during the vaccination period and the inability to obtain antiserum, and considering the amount of serum that needs to be obtained, the number of koi inoculated s...

Abstract

The invention relates to a preparation method of antiserum, and in particular to a preparation method and application of Koi herpes virus antiserum. The preparation method comprises the following steps: selecting healthy Koi as an experimental animal, collecting purified Koi herpes virus antigens, subcutaneously injecting at multiple points of the back of the Koi, when a serum titer reaches a peak value, sampling a great amount of blood from heart or caudal veins, separating serum, measuring a titer, split charging, freezing, and storing. The obtained Koi herpes virus antiserum is used for detecting a KHV (Koi herpes virus) antigen and can be used as positive serum of the KHV antibody detection method. According to the preparation method of the Koi herpes virus antiserum, a preparation method of Koi KHV resistant antiserum is established, the high-titer antiserum is obtained, and the obtained antiserum can be used for detecting the KHV antigen in methods such as IFAT, ELISA and the like and can be used as the positive serum to detect the KHV antibody. A programmed antiserum preparation method is provided for performing the KHV or KHV antibody immunology diagnosis in China, and a finished product immunology diagnosis reagent can be provided.

Description

technical field [0001] The invention relates to a method for preparing antiserum, in particular to a method for preparing koi herpes virus antiserum and its application. Background technique [0002] Koi herpesvirus disease (KHVD) is an infectious and acute viral disease of carp, koi and its variants caused by koi herpesvirus (KHV) infection. The disease infects carp and koi of all ages, causing severe disease and death, with a mortality rate as high as 80% to 90%. Currently, there are at least 22 countries and regions where KHVD occurs in or imported fish, such as Austria, Belgium, Denmark, France, Italy, Luxembourg, the Netherlands, Poland, Sweden, the United Kingdom, China, Hong Kong, Taiwan, Indonesia, Japan, Korea, Malaysia, Singapore and Thailand. KHVD has been reported elsewhere, such as South Africa and the United States. Koi Herpes Virus Disease (KHVD) is listed by the World Organization for Animal Health (OIE) as a fish disease that must be reported, and has iss...
