Embodiment 1

[0032] Embodiment 1: the evaluation of promethazine hydrochloride anti-influenza virus activity

[0033] 1. Experimental Materials

[0034] 1.1 Cells, viruses and drugs

[0035] MDCK cells were purchased from American Type Culture Collection (ATCC); virus strain: A / PuertoRico / 8 / 34 (H1N1); drug: promethazine hydrochloride was purchased from Sigma.

[0036] 1.2 Experimental Instruments

[0037] Multilabel microplate reader (Perkin Elmer).

[0038] 2. Experimental methods and results

[0039] 2.1 Cell culture: 37°C, 5% CO 2 cultured in a humidified incubator. A DMEM medium containing 10% FBS, 100 U / mL of penicillin and streptomycin was used. Cells were subcultured after reaching 90% confluence, and the subculture ratio was 1 / 3 – 1 / 4.

[0040] 2.2 Virus culture: Take SPF chicken embryos aged 9 to 11 days, check them with an egg tester before inoculating the virus, and mark them at a position far away from the embryos, disinfect and punch holes, and inoculate 2- ...

Embodiment 2

[0050] Embodiment 2: Evaluation of the broad-spectrum property of anti-influenza virus effect of promethazine hydrochloride

[0051] In this experiment, promethazine hydrochloride was tested against influenza A subtype H1N1 strain, influenza A virus subtype H3N2 strain, influenza A virus subtype H1N1 amantadine-resistant strain, influenza A virus subtype H6N6 strain, Antiviral activity of influenza virus subtype H7N8 strain, influenza A virus subtype H9N2 strain and influenza B virus.

[0052] 1. The virus strains used in the experiment include: A / Human / Hubei / 1 / 2009(H1N1), A / human / Hubei / 3 / 2005(H3N2), A / human / WSN / 33 / (H1N1,S31N), A / Duck / Hubei / 5 / 2010(H6N6), A / Duck / Hubei / 216 / 1983(H7N8), A / Chicken / Jiangsu / 1 / 2005(H9N2), B / human / Hubei / 1 / 2007.

[0053] 2. Divide MDCK cells into 1.5×10 4 Cells / well were seeded in 96-well cell culture plates, cultured in a 37°C cell culture incubator for 14-18 hours, and the cells were grown into a single layer before use. Discard the medium in t...

Abstract

The invention discloses an application of promethazine hydrochlorine in preparation of a drug for prevention or treatment of influenza virus infection. Firstly, the toxicity of promethazine hydrochlorine on MDCK cells is detected, and a result shows that the maximum non-toxic concentration of promethazine hydrochlorine is 25.0 [mu]M; secondly, the antiviral activity of promethazine hydrochlorine is determined in a completely non-toxic concentration range, and a result shows that the small molecular compound has significant antiviral activity, and the antiviral function has a dose-dependent effect; and finally, the antiviral activity of promethazine hydrochlorine to different types and subtypes of influenza viruses is detected, and a result shows that promethazine hydrochlorine can inhibit replication of all detected influenza viral strains in a dose-dependent mode, thereby representing that the anti-influenza virus activity of promethazine hydrochlorine has a broad-spectrum property. Therefore, promethazine hydrochlorine disclosed by the invention can be used as a novel broad-spectrum anti-influenza virus drug, and can be used for preparation of the drug for prevention or treatment of influenza virus infection.

Description

technical field [0001] The invention relates to the application of promethazine hydrochloride in the preparation of medicines for treating or preventing influenza virus infection, and belongs to the technical field of medicine. Background technique [0002] Influenza viruses belong to the family Orthomyxoviridae, the genus Influenzavirus. According to the differences in the antigenic and genetic characteristics of the virion nucleoprotein (NP) and matrix protein (M), influenza viruses are divided into three types: A, B, and C, also known as A, B, and C. The whole genome of influenza A virus consists of eight single-stranded negative-strand RNAs of different sizes, named after segment 1 to segment 8, respectively. The full-length viral genome is about 13.6 kb, encoding 10 structural proteins (PB2, PB1, PA, HA, NP, NA, M1, M2, PB1-F2 and NS2 / NEP) and non-structural proteins (NS1). Influenza A viruses can be further divided into 17 H (H1-H17) and 10 N (N1-N10) subtypes accor...
