Embodiment 1

[0024] Example 1: Preparation of Methane-containing Physiological Saline Injection

[0025] 1. Degassing treatment. Put a 500ml soft package bag of physiological saline injection in a low-pressure chamber, and treat it under reduced pressure at 0.4 absolute atmospheric pressure for 2 hours at room temperature. After taking it out, place it at room temperature and pressure for 4 hours, and extract the precipitated gas with a syringe. .

[0026] 2. Low-temperature pretreatment Place the above-mentioned degassed injection soft package in an environment of 2° C. for 2 hours to fully cool under normal pressure.

[0027] 3. Inject methane gas Take out 50ml of physiological saline from the above-mentioned low-temperature treated soft packaging bag, and inject 50ml of methane under normal pressure.

[0028] 4. Pressurization to aid dissolution Put the soft package of the injection solution injected with methane into the pressurized cabin below 5°C and continue to pressurize for 12 ho...

Embodiment 2

[0029] Embodiment 2: animal experiments

[0030] 1. Experimental animals: SD male rats, weighing 220-250 g, purchased from the Experimental Animal Center of Second Military Medical University.

[0031] Take 40 male rats and randomly divide them into 5 groups:

[0032] Normal control group, (Control);

[0033] Ischemia-reperfusion injury group, (IR);

[0034] Ischemia-reperfusion damage 0.5ml / 100g treatment group, (IR+0.5ml);

[0035] Ischemia-reperfusion damage 2ml / 100g treatment group, (IR+2ml);

[0036] Ischemia-reperfusion damage 4ml / 100g treatment group, (IR+4ml).

[0037] Except the normal control group, the rest of the groups were given liver ischemia for 60 minutes and then perfused for 6 hours, and the treatment groups with different doses were treated with different doses of methane saline after the end of ischemia.

[0038] 2. Preparation of liver ischemia-reperfusion model:

[0039] SD rats were anesthetized with chloral hydrate and fixed on the mouse board, a...

Abstract

The invention discloses an application of methane injection in preparation of medicaments for treating ischemia-reperfusion injury, which comprises a preparation method of the methane injection. Animal experiments are performed on the methane injection, and the results prove that the methane injection can be used for obviously reducing the level of alanine transaminase, reducing liver injury and reducing hepatocyte apoptosis against a rat model with liver ischemia-reperfusion injury, so that the methane injection can be used for treating the liver ischemia-reperfusion injury. Thus, the methane injection disclosed by the invention can be used for preparing the medicaments for treating the ischemia-reperfusion injury.

Description

technical field [0001] The invention relates to the technical field of medicine, and relates to a methane-containing injection that can be used to prepare medicines for treating ischemia-reperfusion injury. Background technique [0002] Ischemia-reperfusion injury is an important pathophysiological process of cardiac arrest and other diseases. After ischemia-reperfusion, neutrophils and mast cells were infiltrated and activated in the local tissue in the early stage, and a large number of inflammatory factors were produced; in the later stage, there were typical changes such as monocyte infiltration, microglia activation, and astrocyte proliferation. Liver ischemia-reperfusion is a type of tissue ischemia-reperfusion, and the protection of liver ischemia-reperfusion may also suggest that it has a protective effect on other organs from ischemia-reperfusion injury. [0003] At present, the commonly used methods for the treatment of ischemia-reperfusion injury are mainly to re...
