Problems solved by technology

The presence of endogenous substances interferes too much with the determination; second, the above-mentioned trace elements are ubiquitous in natural environments such as air, and it is very easy to introduce exogenous pollution during a series of operations in the experiment
The traditional FAAS method and ICP-AES method can only measure the total amount of elements, and endogenous interference cannot be ruled out, which seriously affects the accuracy of the experimental results

Embodiment 2

[0155] Example 2: In vivo in rats 58 Pharmacokinetic study of Fe-labeled hemin

[0156] There are two main ways to obtain labeled iron compounds: endogenous and exogenous labels. Endogenous labeling refers to the addition of isotopes to feed or cell culture fluid, and the organism itself synthesizes the labeled target compound. This method was often based on radioactive labeling, and it is not ethical to conduct similar experiments in children and pregnant women. At the same time, the method of obtaining endogenously labeled compounds has certain disadvantages. First, the resulting marker abundance is relatively low. In Paz Etcheverry's study, although the preparation contained 58 The hemoglobin of Fe is 30-40 times the content in natural samples, but its abundance only accounts for 9.5%. In addition, iron exists in various chemical forms in the body, such as hemoglobin, ferritin, transferrin, etc. Therefore, the true form of the research object has not been confirm...

Embodiment 3

[0267] Example 3: Mechanism of Unlabeled Hemin Absorption

[0268] initial research

[0269] It is generally believed that the absorption of heme iron is relatively stable and not easily affected by external factors. In contrast, the absorption of non-heme iron is easily influenced by external factors. Diego Moretti et al. found that iron status and food matrix will seriously affect the relative bioavailability of iron pyrophosphate in the human body.

[0270] 1. Fortified iron carrier

[0271] For heme iron, the literature generally reports that there are few factors affecting its absorption. However, some literatures believe that meat can promote iron absorption to a certain extent; some studies have found that zinc can also promote iron absorption. Qianyi Ma et al. utilize radioactive 55 Fe investigated whether bioactive phenolic acids in the regular diet affect iron utilization at the cellular level of Caco-2. Both epigallocatechin and grape seed extract were fou...

Abstract

The invention relates to an application of inductively coupled plasma mass spectrometry in drug testing of hemin. A method for measuring concentration of stable isotope iron in animal plasma is carried out by an ICP-MS (Inductively Coupled Plasma Mass Spectrometry) method and comprises the following steps of: providing an inductively coupled plasma source mass spectrometer; determining operating conditions of the ICP-MS; providing stable isotope iron powder, marked hemin, internal standard elements and the like; preparing standard solution and internal standard solution; preparing ICP-MS diluent; establishing a standard curve; measuring the stable isotope iron concentration in the plasma; and adding the ICP-MS diluent into a sample tube containing animal plasma sample containing iron, mixing uniformly and putting the sample tube into a refrigerator to be refrigerated, and determining the concentration of the iron in the plasma by the obtained standard curve. The application of the inductively coupled plasma mass spectrometry in the drug testing of the hemin has well performances, e.g., the method has well quantification lower limit, accuracy, absolute recovery test, sample stability, medium effect and quality control requirement, and can be taken as a content measurement method for analyzing the content of the stable isotope iron in a biological sample such as the plasma.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and in particular relates to the application of inductively coupled plasma mass spectrometry in the detection of hemin drugs. Background technique [0002] Iron is a very important metal element and an essential trace element for the human body. Iron deficiency is the most common micronutrient deficiency worldwide, affecting 2 billion people including adults, children, and pregnant women. Because of the prevalence and severity of iron deficiency, many countries require iron supplementation to be included in the daily diet. The iron in the current study was mainly non-heme iron preparations, which appeared in the diet in the form of fortified iron. However, heme iron is a kind of biological iron, which will not cause gastrointestinal irritation symptoms, has a high absorption rate, has few factors affecting absorption, and is easy to obtain. It is an ideal iron supplement. Hemin is a stable...
