Embodiment 1

[0061] 1. Design of animal experiments

[0062] Table 1 Animal grouping and dosage

[0063]

[0064] SD rats (body weight 180-210g) were selected as experimental animals, half male and half male, provided by the Department of Experimental Animal Science, Peking University Health Science Center, certificate number: SCXK (Beijing) 2002-0001. After feeding for 2 days, 48 rats were randomly divided into 6 groups, with 8 rats in each group, half male and half male, as shown in Table 1. Feeding method: free intake of feed; water supply method: tap water, animals freely ingested from drinking bottles.

[0065] 2. Sample collection

[0066] Fasting without food and water for 12 hours before administration, administration by intragastric administration according to different concentrations and equal volumes. After 2 hours of administration, the animals were dissected, and blood from the hepatic portal vein (for metabolome) and heart (for genome) were collected. The blood sampl...

Embodiment 2

[0067] Example 2 ECG measurement results

[0068] 1. Heart rate

[0069] Select relatively stable 15-30 seconds from the electrocardiogram of each rat for analysis, and average the data within the selected time to obtain the heart rate of the rat after each administration. Then calculate the average heart rate for the data of the same group at the same administration time. The average heart rate before the administration is recorded as 1, and the average heart rate after 0.5h, 1h, 1.5h, and 2h after the administration is used to compare the heart rate before the administration. The specific analysis results are shown in the appendix figure 1 .

[0070] It can be seen from the above figure that, except for the Shexiang Baoxin Pill group, the heart rate of the other groups decreased to varying degrees after administration, and the degree of heart rate decrease became more obvious with the increase of the dose. The efficacy of the drug starts immediately after administratio...

Embodiment 3

[0078] Example 3 Metabolomics analysis

[0079] 1. Establishment of metabolic fingerprint

[0080] Blood samples were analyzed using a Waters AcquityTM ultra-high performance liquid chromatography system (Waters, Millford, MA) equipped with a high-pressure binary gradient pump, a temperature-controlled autosampler (minimum 4°C) and a diode array detector. Mass spectrometry was performed using a Waters Premier TOF time-of-flight mass spectrometer (Waters, Millford, MA), equipped with an ESI ionization source interface and a Lock-spray interface. A representative rat serum UPLC / TOF-MS metabolic fingerprint is obtained as attached figure 2 shown. The mass spectrometry data was processed by Masslynx software from Waters Company (Waters, MA, USA) for automatic chromatographic peak identification and peak matching, and the spectra obtained by Masslynx were imported into Simca-P, and clustered by the PLS-DA multidimensional statistical analysis system.

[0081] 2.PLS-DA analysi...

Abstract

The invention provides an application of a toad cake to preparation of a medicament for suppressing biosynthesis of oleic acid (OA) and linoleic acid (LA). In a toad cake administration group animal, the metabolic level of AA (Arachidonic Acid) rises, and the metabolic levels of LA and OA fall, which indicate that the toxic and side effects of the toad cake can be used for suppressing the biosynthesis of LA and OA. Metabolic disorder of AA greatly influences normal functions of a heart, can cause reduction in the contraction force of the heart and reduction in cardiac output and can even induce or aggravate myocardial infarct and angina pectoris. By suppressing the biosynthesis of LA and OA through the toad cake, the content of the toad cake in a medicament can be adjusted reasonably, and heart diseases can be better treated or prevented.

Description

technical field [0001] The invention belongs to the field of traditional Chinese medicine, and in particular relates to the application of toad venom in the preparation of medicines for inhibiting the biosynthesis of oleic acid and linoleic acid. Background technique [0002] Chemical Genomics (Chemogenomics or Chemogenomics), as a new technology in the post-genomics era, is a bridge and link between genomics and drug discovery, and will become a powerful tool for functional genomics research. Chemical genomics technology integrates related technologies in the fields of combinatorial chemistry, genomics, proteomics, molecular biology, and pharmacology, and uses biologically active chemical small molecule ligands as probes to study genes closely related to human diseases , the biological functions of proteins, and at the same time provide high-affinity drug lead compounds for the development of new drugs, which is a new model of drug discovery in the post-genomics era and wil...
