Embodiment 1

[0034] A new method for constructing an animal model of brain metastases via carotid artery injection, such as figure 1 shown, including the following steps:

[0035] 1. Research objects: nod-scid mice aged 6-10 weeks were selected.

[0036] 2. Tumor cell preparation: Construct PC90GFP-Luc or A549-GFP-Luc cell lines in advance, passage 2-3 times after recovery, digest tumor cells with trypsin on the day of tumor inoculation, count cells, and divide tumor cells with Hanks buffer Dilute to 2*10 5 / mL and kept on ice for later use.

[0037] 3. Anesthesia: Mice were anesthetized with isoflurane gas.

[0038] 4. Body position and incision:

[0039] A) Exposure of the common carotid artery and common carotid artery bifurcation in the mouse neck:

[0040] Depilate the neck of the mouse, sterilize it with povidone iodine, adopt a longitudinal median incision, about 1 cm in length, retract the skin, expose the salivary glands, retract the salivary glands, expose the trachea, and i...

Abstract

The invention discloses a novel construction method of a cerebral metastatic tumor animal model through carotid artery injection, and relates to the field of animal model construction methods. Comprising the following steps: 1) permanent ligation of the external carotid artery; (2) temporarily blocking the proximal end of the common carotid artery; 3) injecting tumor cells into the common carotid artery; 4) temporarily blocking the telecentric end of the common carotid artery to prevent blood backflow; 5) repairing an injection puncture point with hydrogel; and 6) sequentially loosening the blocking of the telecentric end and the proximal end of the common carotid artery. According to the modeling method, the needed time is short, the technical means difficulty is low, the internal carotid artery is kept unobstructed in the mode of ligating the external carotid artery, intracranial hemodynamics is not obviously changed, the death rate and the postoperative cerebral infarction rate caused by the traditional modeling process are greatly reduced, and the modeling method is suitable for large-scale popularization and application. The process is closer to the process that peripheral tumors enter the encephalic through the internal carotid artery and are colonized in the natural state.

Description

technical field [0001] The invention relates to the field of animal model construction, in particular to a novel method for constructing an animal model of brain metastases via carotid artery injection. Background technique [0002] Brain metastases (BMs) are the most common malignant brain tumors, 10 times more common than primary brain tumors. Among them, it is the main cause of death of cancer patients, accounting for 10%-20% of all cancer patients. Compared with primary tumors, the biological mechanism of brain metastases is still unclear, and the research on brain metastases is still full of challenges. One of the important reasons is the lack of suitable experimental models to accurately simulate the process of brain metastases. Currently widely used preclinical animal models of brain metastases include tail vein injection, cardiac injection, intracranial orthotopic injection, and carotid artery injection. Each model has its advantages and disadvantages, and which mo...
