Abstract

The invention discloses a method for extracting sea cucumber gonad polysaccharide by using microbial protease. The method is characterized by comprising the following steps of: pretreating low-cost sea cucumber gonad serving as a raw material to obtain sea cucumber gonad powder, degreasing and decoloring the sea cucumber gonad powder, dissolving the sea cucumber gonad powder in a phosphate buffer solution according to a solid-liquid ratio of 1: 40, adjusting the pH value of the obtained solution to 7.5, adding the microbial protease, performing enzymolysis at 50 DEG C for 6 hours, sequentially using quaternary ammonium salt and ethanol to precipitate polysaccharide after enzyme deactivation, conducting centrifuging, dissolving a precipitate, performing dialyzing by using a 10-kDa dialysis bag, and carrying out freeze-drying to obtain the sea cucumber gonad polysaccharide. According to the method, the extraction rate of sea cucumber gonad polysaccharide is increased by screening enzymolysis conditions.
