Problems solved by technology

[0004] At present, there are formaldehyde-free preservation solutions on the market, but there is no preservation solution that can simultaneously meet the four screening methods of thin-layer liquid-based cytology detection, HPV detection, immunocytochemical staining and reduced blue histiocytic staining.

Abstract

The invention belongs to the technical field of biology, and discloses a cell preservation solution as well as a preparation method and application thereof. The cell preservation solution comprises alcohol, acid, buffer salt, an ionic strength maintaining agent, an antioxidant and proteolytic enzyme. According to the cell preservation solution, samples of multiple mucus and blood cells can be well treated, and the staining effect of thin-layer liquid-based cytology detection is improved; however, the proteolytic enzyme can influence the staining effect of reductive blue tissue cells, the influence of the proteolytic enzyme on the staining of the reductive blue tissue cells is reduced by adding the antioxidant, and meanwhile, the accuracy and the stability of immune cell chemical staining and reductive blue tissue cell staining are improved; and moreover, the cell preservation solution has no influence on the accuracy and stability of HPV detection after preserving a cell sample. The cell preservation solution can meet requirements of four screening methods including thin-layer liquid-based cytology detection, HPV detection, immunocytochemical staining and vat blue tissue cell staining at the same time.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a cell preservation solution and its preparation method and application. Background technique [0002] Currently, cervical cancer is one of the most common malignant tumors among female cancers. Early diagnosis of cervical cancer and precancerous lesions is the key to improving the cure rate of cervical cancer and the survival rate of patients. Thin-Cytologic Test (TCT) is currently the most widely used clinical method for screening cervical cancer . Thin-layer liquid-based cytology detection is a discipline that diagnoses clinical diseases by observing changes in cell morphology and structure. For decades, thin-layer liquid-based cytology detection has become the main method for large-scale cervical cancer screening and follow-up observation of high-risk groups, which has greatly reduced the incidence of advanced cervical cancer. The smear made by the traditional cyto...
