Embodiment 1

[0028] The preparation method of the probiotic microcapsule based on the complex coacervation of the whole water phase comprises the following steps:

[0029] Step 1. Cultivate the frozen-preserved probiotics, centrifuge, discard the supernatant, and obtain 1×10 9 CFU / ml bacteria slime, spare;

[0030] Step 2. Weigh a certain amount of type A gelatin (GE) powder and dissolve it in ultrapure water to prepare a GE solution with a concentration of 2% (w / w), and keep it warm at 40°C for later use;

[0031] Step 3. Weigh a certain amount of sodium caseinate (NaCaS) powder and dissolve it in ultrapure water to prepare a NaCaS solution with a concentration of 2% (w / w), and keep it warm at 40°C for later use;

[0032] Step 4. At 40°C, 0.3ml of bacterial suspension (1×10 9 CFU / ml), 40g 2% (w / w) GE solution, 20g 2% (w / w) NaCaS solution, 1.2g sucrose (final concentration 2% w / w) powder were mixed, and mixed at 400rpm for 20min; Adjust the pH to 5.5 with % acetic acid, and keep stirrin...

Embodiment 2

[0034] This embodiment is the same as embodiment 1 except that the pH value of mixing coagulation is 5.6.

Embodiment 3

[0036] This embodiment is the same as embodiment 1 except that the pH value of mixing coagulation is 5.8.

Abstract

The invention discloses probiotic microcapsules based on total-aqueous phase composite coagulation and a preparation method of the probiotic microcapsules. The method comprises the steps of (1) obtaining probiotic mud; (2) separately preparing an A-type solution of which the mass concentration is 0.5-5% and a sodium caseinate solution, and performing heat insulation at 30-60 DEG C for standby application; and (3) at 30-60 DEG C, after the probiotic mud, the A type gelation solution and the sodium caseinate solution are mixed, regulating the pH, and inducing a composite coagulation reaction toprepare hygrometric state microcapsules, wherein the addition amount of the probiotic mud lies in that 10<8>cfu-10<12>cfu of the probiotic mud is added to each 1g of the gelatin, the mass ratio of theA type gelatin to the sodium caseinate is (0.25-4) to 1, and the pH range is 5.5-6.0. The composite coagulation based on an A type gelatin / sodium caseinate system does not need an emulsifying step.The method is simplified, the materials are saved, the preparation time of the microcapsules is shortened, and the entrapping rate and storage motility rate of probiotics are increased.

Description

technical field [0001] The invention relates to the fields of food biotechnology and medical technology, in particular to a probiotic microcapsule based on full water phase composite coagulation and a preparation method thereof. Background technique [0002] Probiotics are the most important functional food components. They have the functions of maintaining the balance of intestinal flora, enhancing the body's immunity, controlling cholesterol, blood lipids and blood pressure, and are widely used in foods such as fermented products, dairy products and milk drinks. Only when a sufficient number of viable bacteria reach the human intestinal tract, probiotics can exert their beneficial function. However, probiotics are highly sensitive to environmental factors such as temperature, humidity, oxygen, and acid, and are prone to death during processing, storage, and digestion. , microencapsulation is often used as an important means to improve this problem. The microencapsulation ...
