Embodiment 1

[0025] Embodiment 1. preparation compound 1-16, namely Aspermytin A, Aspermytin A acetate and Phoketide A~N

[0026] 1. Preparation of Total Crude Extract

[0027] 12L of the fermentation liquid of the strain was subjected to conventional ethyl acetate ultrasonic extraction, and the extract was concentrated under reduced pressure to obtain a total crude extract of 7.8 g.

[0028] 2. Separation and purification

[0029] The extract was separated by normal phase silica gel chromatography (200-300 mesh), followed by petroleum ether: ethyl acetate = 80:1, 50:1, 40:1, 30:1, 20:1, 10:1, 5 :1, 1:1, 100% ethyl acetate, and 100% methanol eluent for gradient elution, according to thin-layer plate monitoring, according to the difference in polarity of each fraction, a total of 15 partial elutions were collected from small to large liquid, expressed as Fraction 1-Fraction 15 (Fr 1-Fr 15).

[0030] Fraction 12 (Fr 12) was passed through Sephadex LH-20 gel column (mobile phase was CH 2 ...

Embodiment 2

[0050] Example 2. T cell subtype regulation experiment of the compound of the present invention

[0051] The compounds Aspermytin A, Aspermytin A acetate and Phomaketide A-N of the present invention were tested for T cell subtype regulation by flow cytometry.

[0052] 1. Experimental animal strains

[0053] Female C57BL / 6 mice (18-20 g) were purchased from Shanghai Slack Experimental Animal Co., Ltd. All experimental operations were carried out in strict accordance with the national regulations on the use and maintenance of experimental animals, in line with the guidelines established by the experimental animal center of the author's unit, and approved by the Animal Ethics Committee of the Second Military Medical University.

Abstract

The invention relates to a decahydronaphthalene compound and an application thereof. According to the invention, the decahydronaphthalene compound is separated and extracted from a fermentation liquidof a symbiotic and endophytic fungus Phoma sp. of Sinularia sandensis, wherein the decahydronaphthalene compound has T-cell subtype regulation activity, is low in cytotoxicity and can be used for preparing immunomodulatory drugs. The invention provides a lead compound for developing novel immunoregulatory drugs, opens up a new way for deep research and development of the novel immunoregulatory drugs, and facilitates development and utilization of marine medicinal resources.

Description

technical field [0001] The invention relates to the technical field of medicine, in particular to decahydronaphthalene compounds and applications thereof. Background technique [0002] The host coral Sinularia sandensis belongs to the order Alcyonacea, the suborder Alcyoniina, the family Alcyoniidae, and the genus Sinularia. There are few studies on the soft coral Sinularia sandensis, only 3 reports in the literature, and no report on the chemical composition of its symbiotic microorganisms. At present, there are about 20 compounds isolated from this soft coral, mainly cembrane-type diterpenes (Anjaneyulu, A.; Rao, G.V.; Sagar, K.; Kumar, K.R.; Mohan, K.C., Sandensolide: A newdihydroxycembranolide from the soft coral, Sinularia sandensis Verseveldt of the Indian Ocean.
