Embodiment 1

[0030] Embodiment 1: The effect of berberine on the activity of chemotherapy drug (OXA) peripheral neuropathy cell model

[0031] Instruments: TECAN SPARK10M microplate reader, Olympus IX71 fluorescence microscope.

[0032] Reagents: polylysine, paclitaxel, oxaliplatin and trypsin were from Sigma; collagenase was provided by Worthington; medium, serum, double-antibiotic penicillin and streptomycin were all from Gibco; MTT powder was purchased from Beijing Suolaibao Company; CCK-8 was purchased from Dongren Bio; ATP kit was provided by Lonza Company of the United States; antibodies were purchased from Abcam Company. Both HT-29 cells and SUM-159 cells were donated to other laboratories.

[0033] Cell culture, modeling and drug administration grouping: (1) Isolate and extract the dorsal root ganglion of newborn SD rats, and put them into L15 medium containing 1% double antibody (both 100 units / mL). The supernatant was discarded by centrifugation, and 3mg / mL collagenase was adde...

Embodiment 2

[0037] Example 2: Effect of Berberine on Axonal Growth of Chemotherapeutic Drug (OXA) Peripheral Neuropathy Cell Model

[0038] Experimental apparatus and reagent are the same as embodiment 1.

[0039] (1) Adopt the cell culture method of embodiment 1, the cell suspension is adjusted cell density to be 5 * 10 4 / mL, 500 μL per well was inoculated in a 24-well plate and cultured for 24 hours.

[0040] (2) 30 mM berberine stock solution was dissolved in Neurobasal-B27 (serum concentration reduced to 2%) complete medium containing 3 μM oxaliplatin and serially diluted to a final concentration of 3 μM. The old medium was discarded, and the blank group was replaced with a fresh medium of 2% FBS. The model group and the treatment group were added with 3 μM oxaliplatin, while the treatment group was added with prepared 3 μM berberine. Set up 3 duplicate wells in each group, and continue to culture for 48h.

[0041] (3) Fixation: Discard the medium, wash with PBS three times, 10 mi...

Embodiment 3

[0047] Embodiment 3: the effect of berberine on the activity of chemotherapy drug (PTX) peripheral neuropathy cell model

[0048] Experimental apparatus and reagent are the same as embodiment 1.

[0049] Cell culture, modeling and drug administration grouping: (1) Isolate and extract the dorsal root ganglion of newborn SD rats, and put them into L15 medium containing 1% double antibody (both 100 units / mL). The supernatant was discarded by centrifugation, and 3mg / mL collagenase was added to digest at 37°C for 50min, and the cell mixture was obtained after digestion. (2) Add 1 mL of complete medium containing 10% FBS to stop the digestion, centrifuge and discard the supernatant. Wash twice with L15 medium, add Neurobasal-B27 complete medium containing 10ng / mL GDNF and 10% FBS, and gently blow with a pipette to make a cell suspension. (3) Filter the cell suspension with a 70 μm cell strainer, blow off the cells with a pipette tip and inoculate them on a cell culture plate treat...

Abstract

The invention provides a novel purpose of berberine used for preparing a drug for preventing and treating chemotherapy-induced peripheral neuropathy. The berberine is widely used as a high-efficiencylow-toxin broad-spectrum antibacterial clinical drug for treating gastrointestinal inflammation, and long-term administration cannot generate obvious side-effect. Through establishment of an in-vitroDRG (dorsal root ganglion) cell model and an in-vivo mouse model, the control effect of the berberine on the chemotherapy-induced peripheral neuropathy can be comprehensively evaluated, in addition, the research result also reveals that the berberine has no influence for chemotherapy effect which is inhibition capability for cancer cell growth, and provides powerful evidence for clinical popularization of the berberine for preventing and treating the chemotherapy-induced peripheral neuropathy. The invention firstly provides the novel purpose of the berberine used for preventing and treating the chemotherapy-induced peripheral neuropathy, provides important basis for clinical application of the traditional Chinese medicine to preventand treat the chemotherapy-induced peripheral neuropathy, and has deep meaning.

Description

technical field [0001] The invention relates to a new application of berberine, in particular to an application of berberine in the preparation of drugs for preventing and treating chemotherapy-induced peripheral neuropathy. Background technique [0002] Chemotherapy-induced peripheral neuropathy (CIPN) is a common chemotherapy-related dose-dependent side effect, which is very common in clinical practice, especially in the first month of chemotherapy treatment. Symptoms of CIPN are mostly symmetrical and occur on the sensory, motor and autonomic nerves, but especially sensory nerve side effects, such as skin paresthesia, numbness, tingling, burning, hyperalgesia, tendon reflex disappearance, Loss of vibration sensation and proprioception etc. CIPN not only seriously affects the health and life of patients, but also limits the implementation of chemotherapy regimens and aggravates the condition. However, in 2014, ASCO issued new guidelines for the prevention and treatment o...
