Problems solved by technology

When mass spectrometry is used for protein glycosylation analysis, due to the low content of glycopeptides in biological samples, strong heterogeneity of glycosylation sites, and the presence of a large number of non-glycopeptides that inhibit and interfere with glycopeptide mass spectrometry signals, the analysis results are often unsatisfactory.

Embodiment 1

[0016] (1) Preparation of porous epoxy resin

[0017] Prepare 1 L of an aqueous solution containing 10 g / L of polyvinyl alcohol and 20 g / L of gelatin as the aqueous phase solution of the epoxy resin reaction system prepared by suspension polymerization. Dissolve 120 mg of benzoyl peroxide in a mixed solution containing 12 mL of glycidyl methacrylate, 3 mL of trimethylolpropane trimethacrylate and 18 mL of n-heptane, and add the oily mixed solution to 60 mL of the aqueous phase solution In the process, the obtained oil-water two-phase layered mixed system was stirred at room temperature with a mechanical stirring speed of 300r.p.m. for 1 hour, then the temperature was raised to 80°C, and the polymerization reaction was carried out for 20 hours, and the obtained product epoxy resin was used successively After washing with acetone and ethanol three times, vacuum-dry at 60°C for 12 hours before use.

Abstract

The invention more specifically relates to a method for preparing a hydrophilic resin through a multilayer self-assembly method. The hydrophilic resin can be used for enriching glycopeptides. A porousepoxy resin with the particle size in a range of 100-500 [mu]m is prepared through a suspension polymerization method and is taken as a resin adsorbent, and then, multilayer polysaccharide (hyaluronic acid and chitosan) coats the surface of the porous epoxy resin through a multilayer self-assembly method to prepare the hydrophilic resin. A standard glycoprotein enzymatic hydrolysate is selected as a sample to study the enrichment performance of the hydrophilic resin to glycopeptides. The hydrophilic resin is further applied to glycoproteomics analysis of protein that is extracted from the ratliver.

Description

technical field [0001] The invention specifically relates to a method for preparing a hydrophilic resin by a multilayer self-assembly method, and the hydrophilic resin can be used for glycopeptide enrichment in the field of life and health. Background technique [0002] Protein glycosylation, as an important post-translational modification, regulates many physiological processes. When mass spectrometry is used for protein glycosylation analysis, due to the low content of glycopeptides in biological samples, strong heterogeneity of glycosylation sites, and the existence of a large number of non-glycopeptides that inhibit and interfere with glycopeptide mass spectrometry signals, the analysis results are often unsatisfactory. Therefore, pre-enrichment techniques are required for sample pretreatment before mass spectrometry analysis. At present, a variety of methods including lectin affinity chromatography, phenylboronic acid affinity chromatography, hydrazide chemical method,...
