Embodiment 1

[0038] 1. Activation of strains

[0039] Use YM medium to activate the Geomyces pannorum strain, add 50mL of YM medium to a 250mL conical flask, sterilize the medium with a high-pressure steam sterilizer at 121°C and 0.1MPa for 15 minutes, and then cool to room temperature . The bacteria were inoculated into the Erlenmeyer flask containing the medium, and the inoculation amount was 2% (1 mL) of the fermentation broth. Under the condition of 180rpm and 25°C, the shake flask was activated for 3 days, and then the activation was stopped.

[0040] YM medium: yeast extract 3g, wort juice 3g, peptone 5g, glucose 20g, distilled water 1L.

[0041] 2. Strain fermentation

[0042] After the activation, the modified Chase medium was used for fermentation. Add 50 mL of modified Chapeauer's medium to a 250 mL conical flask, sterilize the medium at 121 °C and 0.1 MPa for 15 minutes with a high-pressure steam sterilizer, and then cool to room temperature. Inoculate the strains into the ...

Embodiment 2

[0067] With compound 1-6 obtained in embodiment 1, each get 1mg after dissolving with the DMSO of 0.5mL respectively, add water for injection as usual, fine filter, potting and sterilizing to make injection.

Embodiment 3

[0069] Take 1 mg of each compound 1-6 prepared in Example 1 and dissolve them in 0.5 mL of DMSO, add water for injection as usual, dissolve it in sterile water for injection, stir to dissolve, and filter with sterile suction Filter through a funnel, pack in ampoules, freeze-dry at a low temperature, and then aseptically melt seal to obtain a powder for injection.

Abstract

The invention discloses a preparation method and application of several thrombolytic compounds generated by polar marine microorganisms. The preparation method comprises the following steps: activating Geomyces pannorum bacteria by using a YM culture medium; fermenting the activated bacteria by using an improved Czapek-Dox culture medium; adding methyl alcohol with volume equal to that of the improved Czapek-Dox culture medium, carrying out ultrasonic treatment, then carrying out centrifugal separation, taking supernatant liquid, and carrying out vacuum concentration and vacuum drying; and carrying out gradient elution of the obtained dry solids through silica-gel column chromatography to respectively obtain compounds 1-6. The compounds 1-6 show thrombolytic activities; EC50 values of evaluation systems are 300, 120.0, 181.3, 300, 200.0 and 179.7 microgrammes per microliter respectively; the compounds 1-6 with the thrombolytic activities can be used as thrombolytic drugs for treating or preventing thromboembolic diseases.

Description

technical field [0001] The invention relates to the preparation method and application of several thrombolytic compounds produced by polar marine microorganisms. Background technique [0002] Thrombotic disease is one of the three major diseases that cause human death, which seriously threatens human life and health. The number of deaths caused by thrombotic diseases has exceeded that of malignant tumors. Thrombolytic therapy is an effective treatment for early acute myocardial infarction and thromboembolic disease. Fibrinolytic therapy is an effective treatment for early acute myocardial infarction and other thrombotic diseases. This therapy uses drugs to activate the potential active plasminogen in the blood to form active plasmin to degrade thrombus fibrin degradation products and dissolve thrombus. . Fibrinolytic therapy is known as one of the top ten discoveries in the field of cardiovascular and cerebrovascular pharmacology in the 20th century. [0003] There are ne...
