Problems solved by technology

Among them, although the microbiological method has the characteristics of simplicity and low cost, it has problems such as time-consuming operation, poor sensitivity and specificity, and cannot meet the needs of current residue detection.
Instrument analysis methods mainly include gas chromatography (GC), high performance liquid chromatography (HPLC) and liquid chromatography-mass spectrometry (LC-MS), etc. These methods have the advantages of accuracy, good specificity, and simultaneous determination of multiple drugs. Requires complex, expensive instruments and equipment, complicated operation, which limits its wide application, and is not suitable for on-site large-scale screening and detection
The conventional enzyme-linked immunosorbent assay (Enzyme-linked Immunosorbent Assay, ELISA) method has the advantages of high sensitivity, high specificity, easy operation, and is suitable for large-scale sample detection, and has been highly valued. However, the existing ELISA is mainly based on Biological antibody and color reaction, among which, biological antibody has the disadvantages of cumbersome preparation, long cycle, poor stability and low sensitivity of color reaction

Embodiment Construction

[0024] The present invention will be further described in detail below in conjunction with the accompanying drawings and embodiments.

[0025] 1. Specific examples

[0026] The present invention uses sulfonamide antibiotic molecular imprinting-quantum dot nanostructure artificial antibody as biomimetic antibody and signal response substance, and establishes a rapid detection method with high sensitivity and selectivity for sulfonamide antibiotics based on the principle of enzyme-linked immunosorbent technology. The specific process as follows:

[0027] 1. Preparation of artificial antibody with molecular imprinting-quantum dot nanostructure

[0028] Add 7.5 mL cyclohexane and 1.8 mL Triton X-100 into a two-necked flask, stir magnetically at 150 rpm for 15 min, then add 400 μL of quantum dot solution with a concentration of 2.5 nmol / mL (quantum dots are CdS / ZnS), 50 μL tetraethyl orthosilicate (TEOS) and 100 μL ammonia water were stirred magnetically for 2 h; then 200 μL temp...

Abstract

The invention discloses a bionic enzyme linked immunosorbent assay method capable of rapidly determining sulfonamide antibiotic residues. The bionic enzyme linked immunosorbent assay method is characterized by comprising the following steps: (1) adding cyclohexane and Triton X-100 into a two-mouth flask; after magnetically stirring, adding quantum dots, tetraethyl orthosilicate and ammonia water in sequence; after magnetically stirring, adding a template molecule solution and a functional monomer; at room temperature, carrying out polymerization reaction to remove sulfonamide antibiotics; dispersing into an ethanol phosphate solution to obtain a molecular imprinting-quantum dot nanostructure artificial antibody solution; (2) adding the molecular imprinting-quantum dot nanostructure artificial antibody solution and a sample extracting solution into each enzyme linked immunosorbent assay hole in sequence; incubating an enzyme linked immunosorbent assay plate at room temperature for 10min and reading a numerical value under an emission wavelength of 605nm by utilizing an enzyme linked immunosorbent assay instrument; calculating the content of the sulfonamide antibiotics in the extracting solution through a standard curve according to a fluorescence value. The bionic enzyme linked immunosorbent assay method has the advantages of high specificity, high sensitivity, simplicity in synthesis, good stability and short detection time.

Description

technical field [0001] The invention relates to a method for detecting residues of sulfonamide antibiotics, in particular to a bionic ELISA detection method for rapidly determining residues of sulfonamide antibiotics. Background technique [0002] Due to the deterioration of aquaculture scale, aquaculture density and aquaculture environment, diseases frequently occur in the aquaculture process, causing serious harm to the development of aquaculture industry. Sulfonamides (SAs) are a class of broad-spectrum antimicrobials that are mainly used for the prevention and treatment of bacterial infectious diseases, or as feed additives for long-term application in animal production. However, irrational use will easily lead to increased drug resistance, and the drug residues in food will cause potential harm to human health, such as inducing cancer. Therefore, its residues in the environment and food have attracted great attention both at home and abroad. Therefore, in order to pro...
