Embodiment 1

[0034] Example 1: Isolation of Lactobacillus plantarum subspecies PS128 and identification of novel strains using ERIC-PCR patterns

[0035] Lactobacillus plantarum subspecies PS128 was isolated from Taiwan's traditional fermented mustard greens - Fucai.

[0036] ERIC-PCR was performed to further differentiate bacterial subspecies with high sequence similarity (Chao et al., 2008; Chao et al., 2009; Rademaker et al., 2007). The ERIC-PCR pattern of the Lactobacillus plantarum strain was carried out with the conditions shown in Table 1 and Table 2. Genomic DNA extracted from PS128 and three Lactobacillus plantarum strains were used as templates for 16S rDNA

Embodiment 2

[0048] Embodiment 2: Identification of Analytical Profile Index (Analytical Profile Index, API)

[0049] The present invention uses the API 50CHL kit (bioMerieux, France) to evaluate the sugar utilization of PS128, and the results are shown in Table 3. Its fermentation test indicated that PS128 has biochemical characteristics similar to Lactobacillus plantarum subspecies.

[0057] (1) Preparation of PS128

[0058] Lactic acid bacteria PS128 were inoculated in culture medium (10% skimmed milk, 1% yeast powder, 0.1% Tween 80, and 2% glucose), cultured at 37° C. for 18 hours, and collected by centrifugation. PS128 was embedded with protective agent (1% skimmed milk, 2% sugar, 2% fructooligosaccharide, 3% maltodextrin and 2% glycerol) and excipients, and freeze-dried to adjust to a final concentration of Powder has 1×10 11 Colony forming unit (colony forming unit, hereinafter referred to as CFU). Store the PS128 powder in a -20°C refrigerator for later use.

Abstract

The present invention provides an isolated lactic acid bacterium, Lactobacillus plantarum subsp. plantarum PS128 deposited under DSMZ Accession No. DSM 28632. The present invention further provides a composition that comprises Lactobacillus plantarum subsp, plantarum PS128 and a carrier. Moreover, the present invention provides a method for prophylaxis or treatment of a stress-induced disorder in a subject that comprises a step of administering an effective amount of Lactobacillus plantarum subsp. plantarum PS128 to the subject. The present invention also provides a method for prophylaxis or treatment of a functional gastrointestinal disorder in a subject that comprises a step of administering an effective amount of Lactobacillus plantarum subsp. plantarum PS128 to the subject.

Description

technical field [0001] The present invention relates to a lactic acid bacterium, in particular to a novel lactic acid bacterium strain for use in the prevention or treatment of stress-induced disorders or functional gastrointestinal disorders in a subject. Background technique [0002] Functional gastrointestinal disorders (FGIDs) are the most common problems in gastroenterology care. These occur as a result of abnormal functioning of the gastrointestinal tract and are defined as chronic abdominal complex syndromes such as abdominal pain, diarrhea, constipation and bloating. According to the Rome Guidelines III, more than 20 functional gastrointestinal disorders have been identified. Common FGIDs include, but are not limited to, functional abdominal pain, irritable bowel syndrome (IBS), constipation, functional diarrhea, and functional dyspepsia. [0003] Irritable bowel disorder is a chronic functional gastrointestinal disorder. About 4% to 30% of people worldwide suffer...
