Abstract

The invention discloses a method for inhibiting a dysosma versipellis explant from contamination and browning. The method comprises the following steps: transplanting a wild dysosma versipellis rhizome which is more than five-year old into clean river sand matrix for preculture; soaking a stem growing strongly into N-long-chain thiocarboxylic acid glycine betaine, washing with distilled water, and brushing the dirt on the surface of the explant; treating the explant by use of anion photocatalysis, sterilizing with sodium hypochlorite, washing with sterile water, sucking moisture on the surface by use of aseptic filter paper until the surface is dry, cutting into squares, and soaking in a heavy metal complexing agent; inoculating onto a modified MS medium for dark culture for two weeks; and then performing bright culture. The method can be used of effectively preventing the dysosma versipellis explant from contamination and browning and providing sufficient aseptic explants for the tissue culture research of the medicinal plant-dysosma versipellis, so that the follow-up tissue culture success rate is ensured, the cost is low and the method is suitable for industrial production.
