Abstract

The invention provides a preparation method for a steelhead trout and arctic char hybrid triploid seed. The preparation method includes the steps that steelhead trout female broodstock egg and arctic char male broodstock sperm are in vitro fertilized; after the fertilized egg is fertilized for 12-14 minutes, the fertilized egg is moved into 6-DMAP (dimethylanminopyridine) solution with concentration of 170-190 milligrams per liter and temperature of 9-11 degrees Celsius to be soaked for 14-16 minutes; and then the soaked fertilized egg is moved into water of the temperature of 6-8 degrees Celsius to be hatched. After about 50 days, a hybrid triploid larvae is hatched by the fertilized egg. During incubation period, eyed rate of the fertilized egg is above 81%, and hatching rate of the seed reaches above 90%. Then the obtained larvae is detected through a flow cytometry, and triploid rate of the obtained larvae reaches 94%. With the preparation method for the steelhead trout and the arctic char hybrid triploid seed, during trout breeding process, problems of precocious puberty, individual miniaturization, disease breeding and the like in an individual fish are alleviated.
