Abstract

The invention discloses a method for preparing an ACE (Angiotensin Converting Enzyme) inhibitory peptide by hydrolyzing rice protein isolate with a complex enzyme. The method comprises the following steps of: (1) adjusting temperature of rice protein isolate aqueous solution with mass-volume concentration of 5-8 percent to 45-55 DEG C and pH (Potential of Hydrogen) value to 8.5-9.5, and adding alkaline protein enzyme according to an enzyme adding amount of 48-60 AU/kg to hydrolyze for 120-150 min; (2) adjusting system temperature to 36-38 DEG C and pH value to 7-8 and adding trypsin according to an enzyme adding amount of 12.5-25 USP/kg to hydrolyze for 30-60 min; and (3) adjusting the pH value to 5.0-5.5 and performing solid-liquid separation, ultrafiltration membrane screening, desalting, concentration and freeze drying on enzymatic hydrolysate in sequence to prepare the ACE inhibitory peptide product.
