Problems solved by technology

[0002] Rhizopus niger (see figure 1 and figure 2 ) Rhizopus stolonifer, also known as stoloniferous rhizopus, belongs to Zygomycetes (Zygomycetes) Mucorales (Mucorales) Zygomycotina (Zygomycotina) fungus, mostly born on bread, steamed buns and foods rich in starch, making food Rotten and deteriorated, the Rhizopus nigricans used in the present invention is separated and purified from the Changbai Mountain wild rubber gyro water infusion, because fungi have many types in microorganisms, many secondary metabolites, and relatively simple culture conditions, etc., so they are the best choice for fermenting Chinese medicines. The main functional bacteria, but the research on the biotransformation of panaxatriol group saponins with it has not been reported

Embodiment Construction

[0023] Preparation of Rhizopus nigricans strains: place Changbai Mountain wild rubber gyro water infusion for a week, pick Rhizopus niger on the water, connect to PDA flat culture medium, keep pH natural, separate and purify for many times, until pure Rhizopus niger is obtained , transferred to PDA slant medium for 3 days, and placed in a 4°C refrigerator for later use.

[0024] Preparation of ginsenoside fermentation product: Accurately weigh 0.500 ± 0.020 g of ginsenoside group saponins and put it into a 250 mL Erlenmeyer flask, put 100 mL of PD medium in each culture bottle, put it into a sterilizer in time for sterilization, and put it in a steam pressure of 11.76 ×104Pa~12.75×104Pa, keep it for 30min, take it out and place it on the ultra-clean workbench. Set at 28°C, rotation speed 125r / min, after 7 days of conversion time, vacuum pump to filter the fermentation broth, extract the filtrate three times with water-saturated n-butanol 1:1, take the upper n-butanol extracts ...

Abstract

The invention discloses a rhizopus stolonifer capable of biologically converting ginsenosides. A fermentation product is analyzed by an HPLC (High Performance Liquid Chromatography) system to prepare a component with a reduced color spectrum peak value in an original panaxatriol saponins and a component with an increased peak value in the panaxatriol saponins fermentation product thereof. By isolation and identification, a converted substance in the panaxatriol saponins is confirmed to be ginsenosides Re, a compound (1) in the panaxatriol saponins fermentation product is confirmed to be ginsenosides Rh1, a compound (2) is confirmed to be ginsenosides Rg3, and a compound (3) is confirmed to be ginsenosides Rg2. By speculation, the ginsenosides Re in the panaxatriol saponins is converted into the ginsenosides Rh1 by the black rhizopus or the ginsenosides Re is firstly converted into the ginsenosides Rg2 and then converted into ginsenosides Rh1, and the detailed conversion path is to be further researched.

Description

Technical field: [0001] The invention discloses a fungus Rhizopus stolonifer that can biotransform ginsenosides. Through HPLC system detection, the result shows that it can make some monomeric saponins inside the ginsenosides A conversion occurs. The invention belongs to the application of microorganisms in the field of biotransformation. Background technique: [0002] Rhizopus niger (see figure 1 and figure 2 ) Rhizopus stolonifer, also known as stoloniferous rhizopus, belongs to Zygomycetes (Zygomycetes) Mucorales (Mucorales) Zygomycotina (Zygomycotina) fungus, mostly born on bread, steamed buns and foods rich in starch, making food Rotten and deteriorated, the Rhizopus nigricans used in the present invention is separated and purified from the Changbai Mountain wild rubber gyro water infusion, because fungi have many types in microorganisms, many secondary metabolites, and relatively simple culture conditions, etc., so they are the best choice for fermenting Chinese me...
