Embodiment 1

[0028] Example 1: Preparation of Rat Adipose Stem Cells (ADSCs)

[0029] Isolation and culture of rat ADSCs

[0030] Rats were anesthetized by intraperitoneal injection of 10% chloral hydrate (3ml / kg), and bilateral inguinal fat pads were taken under aseptic conditions and placed in PBS solution. Wash the fat pad 3 times with PBS, remove the visible blood vessels and fibers, and cut them into pieces. Digest with 0.075% type II collagenase at 37°C for 3Omin (by pipetting once every 10min). An equal volume of DMEM / F12 culture medium containing 10% FBS was used to neutralize the collagenase, centrifuged at 1100r / min for 5min, and the supernatant and adipose tissue were removed. The DMEM / F12 complete culture solution was resuspended, and filtered through a 400-mesh cell sieve (pore size 60 μm). Take 1×10 5 / ml inoculum, 37°C, 5% CO 2 and 95% humidity conditions, and change the medium after 24 hours to remove unattached cells, lipid droplets and residues. Thereafter, the medi...

Embodiment 2

[0042] Example 2: Construction of Tissue-Engineered Breast Grafts

[0043] Construction of Adipose Stem Cells / Type I Collagen Gel Complex (ADSCs-COL)

[0044] Cells were cultured with the above-mentioned adipogenic inducers 24 hours prior to construction of tissue-engineered breast grafts. 10 bottles (25cm 2 Culture flask) The 3rd passage ADSCs cells grown and confluent were digested and centrifuged, and then added to 540 μl of culture medium, containing about 2×10 cells 7 indivual.

[0045] Type I collagen solution: the concentration of type I collagen is 5mg / ml, dissolved in 0.006mol / L acetic acid. Add 400 μl type I collagen solution to 24 μl 0.1 mmol / L NaOH in an ice bath, mix immediately, then add 46 μl 10×PBS, and measure the pH value to be 7 after mixing. Then add the above 540 μl cell suspension to form adipose-derived stem cell / type I collagen gel complex (the final concentration of collagen is 2 mg / ml, and the cell density is 2×10 7 / ml). The solution can be sol...

Abstract

The invention belongs to the technical field of construction of artificial organs by a tissue engineering method in biomedical engineering and relates to a tissue engineered breast transplant and a constructing method thereof. The tissue engineered breast transplant of the invention takes injectable material I type collagen gel as a carrier bracket and takes adipose-derived stem cells as seed cells; the seed cells are attached to the carrier bracket to form an adipose-derived stem cell / I type collagen gel compound; the final concentration of collagen is 2 mg / ml; and the cell density is 2*10<7> / ml. The tissue engineered breast transplant of the invention consists of injectable material-loaded autologous adipose-derived stem cells and is simple and convenient to prepare. Experiments prove that a new adipose tissue can be produced and the tissue engineered breast transplant is finally converted into autologous fat without an immunological rejection problem, so a new solution is provided for the filling and reconstruction of a breast.

Description

technical field [0001] The invention belongs to the technical field of constructing artificial organs by tissue engineering method in biomedical engineering, and in particular relates to a tissue-engineered breast graft and a construction method thereof. Background technique [0002] Due to reasons such as race, heredity and nutrition, a considerable part of women's breasts are dysplastic or loose and atrophied. Breast augmentation is favored by the majority of beauty-loving women for its immediate effect. Breast augmentation is currently used to improve female breast dysplasia or loose atrophy, as well as breast loss due to tumor removal or trauma. In the United States, 200,000 women receive breast augmentation surgery every year. There is no accurate statistics on the number of breast augmentation patients in China every year, but the literature speculates that the number may be between 300,000 and 500,000. At present, the current methods of medical breast augmentation a...
