Problems solved by technology

However, in the current research, the diameter of PHBV electrospun fibers is generally on the micron scale, the degradation rate is not high, it is difficult to match the bone formation rate, and the film-like electrospun fibers lack sufficient strength, so electrospun fibers are used in the research of bone defect repair Still stuck in the stage of in vitro cell experiments

[0013] The preparation method of the degradable three-dimensional fiber scaffold that promotes the repair of bone defect with PHBV as the matrix provided by the present invention comprises the following steps:

[0014] First, the biomaterial polyhydroxybutyrate valeric acid copolyester PHBV, which has good water solubility and can promote the overall degradation rate of the scaffold, polyoxyethylene PEO, and nano-sized hydroxyapatite nHA, which has osteoconductivity but requires a carrier, were selected as raw materials. The PHBV composite nanofiber membrane was prepared by electrospinning, and then cut and folded to obtain a multilayer rod-shaped three-dimensional fiber scaffold, which was used as an implant material for large-scale bone defect repair.

[0015] The degradable nanofiber film is obtained by mixing PHBV and PEO in a certain ratio, dissolving them in trifluoroethanol (TFE) solvent, and realizing by electrospinning. The ratio of PHBV and PEO is selected by preparin...

example

[0022] 1. PHBV was added to the solvent trifluoroethanol (TFE), and stirred with a magnetic stirrer until it was completely dissolved. In order to obtain nano-scale fibers and obtain a degradation rate that matched the bone regeneration rate, another biomaterial, polyoxyethylene ( PEO), the mass ratio of PHBV to PEO is 9:1, 8:2, 7:3, 6:4, 5:5;

[0023] 2. Add nHA to TFE, ultrasonic for 90 minutes, so that nHA is evenly dispersed in TFE, and the amount of nHA added is 10% (mass percentage) of PHBV / PEO;

[0024] 3. Mix and stir the solution obtained in steps 1 and 2 evenly to obtain an electrospinning solution with a final concentration of 2%;

[0025] 4. Add the spinning solution into a 20ml syringe. The needle of the syringe is flat and the inner diameter is 0.5mm. The syringe is fixed on the propulsion pump, and the propulsion speed is set to 5ml / hour. The positive pole of the high voltage DC power supply is connected to the syringe needle, and the negative pole receives (Ro...

Abstract

The invention provides a preparation method for a degradable three dimensional fiber scaffold capable of promoting the repair of bone defects bone defect, and relates to a preparation method for a nano fiber scaffold material based on an electrospinning technique. The preparation method combines a biological material of polyhydroxybutyrate-hydroxyvalerate copolyester (PHBV) which is degradable, has no immunogenicity but cannot induce osteogenesis, polyoxyethylene (PEO) which has good water solubility and can promote the scaffold to be degraded rapidly and hydroxyapatite (nHA) which has bone conductive capacity but need a certain vector. An electrospinning fiber membrane with nano-scale diameter is obtained via an electrostatic spinning method; and finally a multilayer rod-shaped degradable three-dimensional scaffold material can be obtained via cutting and folding. Animal experiments demonstrate that the scaffold has good biocompatibility and appropriate degradable speed, and can realize the repair of large-size bone defects.

Description

technical field [0001] The invention relates to a method for preparing a degradable three-dimensional fiber scaffold for promoting bone defect repair, and belongs to the technical field of tissue engineering. Background technique [0002] Bone is a surprising yet real nanocomposite, a complex and highly differentiated connective tissue. Not only does it provide mechanical support for the body, it is also a source of various minerals for the body, especially calcium and phosphorus. Bone is a representative of a dynamic tissue with unique self-regeneration and remodeling capabilities. Bone can grow to a certain length throughout a person's life without any scarring. Although bone has the ability to regenerate and remodel, its regenerative ability only works when the defect site is small, and large-scale bone reconstruction is still a clinical problem. When the defect is so large that the bone cannot heal itself, then a bone graft is needed to help restore bone function. The...
