Problems solved by technology

Li Defu et al. conducted research on sulfate-reducing bacteria (SRB), first put SO 4 2- reduced to H 2 S, then by H 2 S reduces hexavalent chromium and applies it to the treatment of electroplating wastewater, but the treatment process will produce toxic H 2 S gas is difficult to control, thus limiting the application of this method
Japanese scholars once reported that Enterobacter cloacae HO1 (E.cloacae HO1) was used to treat chromium-containing industrial wastewater under anaerobic conditions. In order to form anaerobic conditions in this method, a large amount of nitrogen must be filled in the medium to remove the medium. dissolved oxygen, which greatly increases operating costs and process operability

[0024] Example 1. Growth and reduction of hexavalent chromium in four culture media of Pallidum pallidum CTS-325

[0025] 1.1 Recovery of strains

[0026]Take the seed bacteria preserved in liquid nitrogen, put them into a 100ml Erlenmeyer flask containing 20ml of 1# liquid medium (pH 7-8) at a ratio of 1:100, and place them at 200 under aerobic, normal pressure, and 30°C conditions Shake the culture overnight in a shaker with rpm to revive the strain.

[0027] 1.2 Cultivation of Paleobacterium pallidus CTS-325 in different media and reduction of hexavalent chromium

[0028] Transfer the resuscitated strains into four 250ml Erlenmeyer flasks containing 100ml of culture medium at a ratio of 1:100 of the culture medium to the culture medium. The composition of the culture medium in the four Erlenmeyer flasks is different. , 1# culture medium (L -1 ): tryptone 10g, yeast extract 5g, sodium chloride 10g pH=7.0~7.5; 2# culture medium (L -1 ): tryptone 10g, beef extract 3g, sodi...

[0029] Example 2. Situation of reduction of hexavalent chromium in 1# culture medium by Paleobacterium pallidus CTS-325

[0030] According to Example 1, we selected the 1# medium with the best growth and reduction effect to carry out the following experiments. Different initial concentrations of hexavalent chromium were added to the medium, which were 100mg / L and 200mg / L respectively.

[0031] 2.1 Recovery of strains

[0032] Take the seed bacteria preserved in liquid nitrogen, put them into a 100ml Erlenmeyer flask containing 20ml of 1# liquid medium (pH 7-8) at a ratio of 1:100, and place it at 200 Shake the culture overnight in a shaker with rpm to revive the strain.

[0033] 2.2 Reduction of hexavalent chromium by Paleobacterium pallidus CTS-325 at different initial chromium concentrations

[0034] Transfer the resuscitated strains into two 250ml Erlenmeyer flasks containing 100ml of 1# medium according to the ratio of bacterial solution to medium 1:100, and the concentr...

[0037] Example 3. The situation of reducing hexavalent chromium in the optimized 1# medium of Paleobacterium pallidus CTS-325

[0038] In order to solve the problem that Paleobacterium pallidum CTS-325 cannot completely reduce hexavalent chromium in 1# medium, we optimized 1# medium. By adding xylose or sucrose to the 1# medium, or the bagasse left over from the production of xylose and sucrose, it was found that the effect of reducing hexavalent chromium in the optimized 1# medium of Pallidum bacillus CTS-325 was significantly improved, and Hexavalent chromium can be completely reduced when the initial hexavalent chromium concentration is 100mg / L and 200mg / L respectively.

Abstract

The invention discloses Ochrobactrum CTS-325 and a culturing method thereof as well as application to reducing hexavalent chromium, and relates to microbe and application thereof to efficiently and thoroughly reducing the hexavalent chromium under the aerobic condition as well as a culturing method thereof. The Ochrobactrum CTS-325 has a preserving number CGMCC1715. The strain can efficiently and thoroughly reduce the hexavalent chromium. The pH value is between 7 and 9 during reduction of the hexavalent chromium by the strain. The strain comprises one or combination of any few kinds of xylose, ribose, glucose, levulose, galactose, sucrose, maltose, starch, glycerin or mannite in an available culture medium on microbiology. When industrial waste water containing chromium and chromium slag leach liquor are treated by the Ochrobactrum CTS-325 in an optimized culture medium, the hexavalent chromium in the industrial waste water and the chromium slag leach liquor can be reduced to reach sewage discharge standard.

Description

technical field [0001] The invention relates to microorganisms and their application and cultivation method for efficient and thorough reduction of hexavalent chromium under aerobic conditions. Background technique [0002] Chromium salt products are indispensable and important chemical raw materials for the development of the national economy, and are widely used in electroplating, printing and dyeing, leather tanning, wood preservation, and oil exploration. In the process of production and application of chromium salt products, a large amount of chromium pollution will inevitably be produced, which seriously threatens human health. According to reports, drinking water contaminated by chromium can cause abdominal discomfort and diarrhea and other poisoning symptoms; chromium is a skin allergen, which can cause allergic dermatitis or eczema; chromium can stimulate and corrode the respiratory tract, causing rhinitis, Pharyngitis, bronchitis, severe nasal septum erosion, and ...
