Embodiment 1

[0085] The effect of a combination of flavonoids and tocotrienols on lowering cholesterol in human subjects was studied.

[0086] Research design :

[0087] For ten hypercholesterolemia patients with serum total cholesterol > 5.9mmol / L, LDL cholesterol 4.0mmol / L and serum triacylglycerol 230mg / dl, >155mg / dl and <307mg / dl) Subjects were given a daily supplement consisting of 270 mg polymethoxylated flavones and 30 mg tocotrienols for four weeks. Participants in this study must be free of thyroid disorders, kidney disorders and diabetes. At the same time, subjects taking cholesterol-lowering drugs were asked to stop the treatment four weeks before the study.

[0088] To determine whether the treatment improved parameters associated with a high risk of heart disease, fasted blood samples were drawn at the beginning of the study and at the end of 4 weeks for analysis of plasma total and lipoprotein cholesterol, plasma apolipoprotein B (related to LDL ) and A1 (associated with...

Embodiment 2

[0097] Clinical Trial 2 : Cholesterol-lowering properties of combinations of polymethoxylated flavonoids and tocotrienols in human subjects

[0098] The purpose of this study was to evaluate the cardioprotective ability of a combination of flavonoids and tocotrienols in human subjects.

[0099] Research design :

[0100] A supplement consisting of 270 mg polymethoxylated flavones and 30 mg tocotrienols was administered daily for four weeks to ten hypercholesterolemic subjects. Participants in this study must be free from thyroid disorders, kidney disorders and diabetes. At the same time, subjects taking cholesterol-lowering drugs were asked to stop the treatment four weeks before the study.

[0101] Blood samples were taken from a forearm vein before initiation (baseline) and at the end of the 4-week period. Plasma lipids profiles and other metabolic parameters were analyzed using standard methods. Seated blood pressure was recorded using a conventional mercury manomet...

Abstract

Disclosed in certain embodiments is a pharmaceutical formulation, such as a tablet, gel capsule, solution, liquid, suspension or emulsion, comprising a pharmaceutical ingredient comprising an active agent combination comprising flavonoids and tocotrienols in a ratio of about 75:25 to about 95:5 and at least one pharmaceutical acceptable excipient. The flavonoids may comprise naringenin, hesperetin, nobiletin or tangeretin. The tocotrienols may comprise alpha-tocotrienol, gamma-tocotrienol or delta-tocotrienol. The formulations may be used to lower total cholesterol, triacylglycerols, LDL cholesterol and Apo B.

Description

[0001] related application [0002] This application claims priority to US Provisional Application No. 60 / 574,490, filed May 26, 2004, which is incorporated herein by reference. Background technique [0003] Hyperlipidemia is a pathological condition in mammals in which abnormally high concentrations of serum circulating lipids are present. The lipid pool components in the circulation are mainly composed of triglycerides (glycerides of fatty acids), cholesterol, and fatty acid esters of cholesterol. It is found that such molecules are usually bound to specific proteins in a complex that serves as a transport mechanism. Hyperlipidemia is a disorder generally associated with elevated levels of cholesterol, phospholipids, and / or triglycerides in the serum of mammals. [0004] Hyperlipidemia includes six heritable forms of hyperlipoproteinemia; these types are often referred to as lipoprotein phenotypes. Most plasma lipids, including cholesterol and triglycerides, do not circul...
