The invention discloses a method for determining the opportunity of inducing 2n pollen of phalaenopsis amabilis by colchicine. The method comprises the following steps of (a) counting the yield of natural 2n pollen; (b) conducting 2n pollen induction: selecting the pollen in each meiosis period, and treating flower buds with 0.01%-0.1% colchicine solution to perform 2n pollen induction; (c) counting the 2n pollen rate after the induction; and (d) comparing the 2n pollen rate obtained after induction with the natural 2n pollen rate so as to determine the opportunity for inducing the 2n pollen. The invention also provides a method for inducing 2n pollen of phalaenopsis amabilis by using the colchicine. According to the method, the opportunity for inducing the 2n pollen by utilizing the colchicine and the corresponding colchicine concentration are determined, and a foundation is laid for obtaining the 2n pollen through polyploid induction and cultivating a new phalaenopsis variety which is large in flower size, bright in color and high in ornamental value.
