Embodiment 1

[0024] 1. Experimental design

[0025] Healthy carp were bred in an indoor circulating water system and domesticated for 2 weeks. During the acclimatization period, the commercial feed was fed twice a day, and the feeding amount was 2% of the fish body weight. The breeding conditions are: temperature 26±3℃, dissolved oxygen>5mg / L, pH 7.3±0.2, ammonia nitrogen content<0.1mg / L, nitrite content<0.1mg / L.

[0026] After domestication, the carps were randomly divided into 4 groups and exposed to 4 different concentrations of H 2 o 2 : 0, 0.25, 0.5 and 1 mM), the 0 mM group was the control group (CK), 60 fish in each group, divided into 3 parallel repetitions. Expose for 1 hour per day for 14 days. After 14 days of exposure, 10 fish were randomly selected from each concentration, anesthetized, and blood and liver tissues were collected. Determination of stress indicators in blood: cortisol, lactic acid and glucose; determination of oxidative stress indicators in liver tissue: ca...

Abstract

The invention discloses a carp chronic oxidative stress model and a construction method thereof. According to the method, hydrogen peroxide is adopted to induce carps in feeding, and the use concentration of the hydrogen peroxide is 0.25-1 mM. Compared with the prior art, the method has the following advantages that (1) the chronic oxidative stress model is constructed in the fish for the first time, the successful construction of the model is beneficial to the research of a chronic oxidative stress injury mechanism of the fish, and meanwhile, the method can be used for screening fish antioxidant drugs; (2) the hydrogen peroxide is adopted to directly induce the fed carps, the model construction mode is simple, and the success rate is high; and (3) the carp sources used in the method are consistent, and the culture environment is controllable, so that the repeatability is strong.

Description

technical field [0001] The invention belongs to the field of aquatic disease prevention and control, and relates to the construction of a stress model, in particular to a carp chronic oxidative stress model and a construction method thereof. Background technique [0002] Under intensive farming conditions, fish are stressed by various stress factors, causing stress responses. In the stress response, oxidative stress is considered to be the most important stress mode. Various factors in the breeding environment, such as high breeding density, unsuitable temperature, and pathogenic microorganism infection, can induce the generation of reactive oxygen species (ROS) in the body and cause oxidative stress. In the state of oxidative stress, excessive ROS will consume antioxidant substances, damage the antioxidant defense system, attack biofilms, oxidize biomacromolecules (proteins, lipids and nucleic acids), cause damage to body tissues, and affect the normal life of fish. Physi...
