preparation example Construction

[0095] Some embodiments of the present application also provide a preparation method of the aforementioned coating solution for improving the stability of Chlamydia pneumoniae antigen / Mycoplasma antigen in immunochromatographic reagents, including:

[0096] Mix the buffer, the first component, stabilizer, antioxidant and preservative evenly;

[0097] Among them, 3ml~5ml first component, 0.3g~1.5g stabilizer, 0.5g~2g antioxidant and 0.02ml~0.04ml preservative are added to every 100ml buffer.

[0098] Further, the concentration of the aforementioned buffer is 10-30 mM. Further optionally, the concentration of the above-mentioned buffer is 12-28 mM. Further optionally, the concentration of the aforementioned buffer is 14-26 mM. Further, the above-mentioned buffer solution is prepared by dissolving 0.145 to 0.435 parts by weight of disodium hydrogen phosphate, 0.012 parts to 0.036 parts by weight of sodium dihydrogen phosphate, and 0.85 parts by weight of sodium chloride in 100 ml of d...

Embodiment 1

[0105] The coating solution for improving the stability of Mycoplasma pneumoniae antigen in immunochromatographic reagents provided in this example is prepared as follows:

[0106] 1) Reagent manufacturers: sodium dihydrogen phosphate, disodium hydrogen phosphate, sodium chloride, and methanol were purchased from Guangzhou Chemical Reagent Factory; trehalose was purchased from Nanjing Zhongnuo Biological Engineering Co., Ltd.; bovine serum albumin, TCEP, EDTA- 2Na + , Proclin300 was purchased from Sigma-Aldrich (Shanghai) Trading Co., Ltd.

[0107] 2) Preparation of Mycoplasma pneumoniae antigen coating solution: Weigh 0.29 g of disodium hydrogen phosphate, 0.024 g of sodium dihydrogen phosphate, and 0.85 g of sodium chloride in 100 ml of deionized water to obtain a molar concentration of 20 mM PBS buffer; in 100 ml Add 5g of trehalose, 3ml of methanol, 0.5g of bovine serum albumin, 2g of TCEP, 2g of EDTA-2Na to the 20mM PBS buffer + , 0.02ml of Proclin300, stir until all the compo...

Embodiment 2

[0109] The basic steps are the same as in Example 1, except that 3 ml of methanol is replaced with 3 ml of isopropanol.

Abstract

The invention relates to the field of biological reagents, in particular to coating liquid for improving the stability of chlamydia pneumoniae antigen / mycoplasma antigen in an immunochromatographic reagent and a preparation method thereof. The coating solution comprises a buffer solution, wherein the buffer solution comprises a first component for immobilizing a chlamydia antigen or a mycoplasma antigen on a nitrocellulose membrane, a stabilizer, an antioxidant and a preservative. The first component is beneficial to immobilization of the antigen on the nitrocellulose membrane. The stabilizercan improve the stability of the mycoplasma antigen and the chlamydia antigen. The antioxidant can prevent the mycoplasma antigen and the chlamydia antigen from being oxidized. The preservative can play a role in preventing mycoplasma antigens and chlamydia antigens from being corroded. All the components in the coating liquid cooperate with one another to protect the stability of the antigen in the immunochromatographic reagent.

Description

Technical field [0001] This application relates to the field of biological reagents, in particular to a coating solution for improving the stability of Chlamydia pneumoniae antigen / Mycoplasma antigen in immunochromatographic reagents and a preparation method thereof. Background technique [0002] Chlamydia pneumoniae and Mycoplasma pneumoniae are common human respiratory tract pathogens, which often cause acute respiratory infections, especially pharyngitis, sinusitis, bronchitis, pneumonia and other diseases. [0003] The detection methods of Chlamydia pneumoniae and Mycoplasma pneumoniae include pathogen isolation and culture method, direct antigen detection method, nucleic acid amplification detection method, serological detection method, enzyme-linked immunosorbent assay (ELISA), immunochromatography, polymerase chain reaction Method (PCR) and so on. Among these detection methods, serological detection methods are the most commonly used detection methods because they are easy ...
