Problems solved by technology

However, although the application of TMZ has significantly improved the therapeutic effect of glioma, it is difficult to pass through the blood-brain barrier, and the effective concentration of the drug is reduced due to the high hydrostatic pressure in the space between the tumor and surrounding tissues. Drug resistance, adverse reactions to the body, and treatment difficulties caused by molecular genetic changes will all have adverse effects on the treatment of glioma.

Embodiment Construction

[0016] Next, the MTT method was used to detect the effect of epetecyl hydrochloride on the growth and proliferation of human glioma cell lines U251 and SHG44 cells, and the effects of genomic DNA electrophoresis, DAPI staining and TUNEL staining on human glial cells cultured in vitro were detected. The apoptosis-inducing effect of tumor cell SHG44 cells is verified by mouse animal experiments. The materials and reagents used in the experiment are as follows:

[0017] 1.1 Materials and methods

[0018] cell line

[0019] Human glioma cell lines U251 and SHG44 were preserved by the Neurosurgery Department of the First Hospital of Jilin University and cultured in DMEM medium containing 10% calf serum at 37°C and 5% CO2.

[0020] 1.2 Main reagents and instruments

[0021] Temozolomide (TMZ), ecetoline hydrochloride (abbreviated as CPL), tetramethylazolazolium blue (MTT), and DAPI were purchased from Sigma; TUNEL cell apoptosis detection kit was purchased from R&D; Triton X-100 w...

Abstract

The invention provides application of cephaeline hydrochloride in preparing medicine for treating glioma, and belongs to the technical field of medicine. The method comprise the steps that an MTT method is adopted for detecting the influence of the cephaeline hydrochloride on growth and proliferation of human glioma cell lines U251 and SHG44; genomic DNA electrophoresis, DAPI staining, TUNEL staining and other methods are adopted for detecting the apoptosis induction effect of the cephaeline hydrochloride on human glioma cell SHG44 cells cultured in vitro. The results show that the cephaelinehydrochloride has the effect of inhibiting proliferation of the glioma cells.

Description

technical field [0001] The invention belongs to the technical field of medicines, and in particular relates to the application of epetolline hydrochloride in the preparation of medicines for treating glioma. Background technique [0002] Glioma is the most common primary intracranial tumor. About 81% of malignant brain tumors are glioma, which is characterized by short course of disease, difficult cure, and high mortality. The survival rate is only 34%. At present, surgical treatment is the first choice for the treatment of this disease, combined with radiotherapy, chemotherapy, immunotherapy, molecular targeted therapy and other methods to form a comprehensive treatment strategy. The treatment of glioma still faces many difficulties. Finding drugs with low toxicity and side effects that can target tumors has always been the goal of anti-tumor drug development. [0003] Since the clinical application of the alkylating agent TMZ (temozolomide) in 2005, great progress has be...
